Aspergillus niger is an efficient cell factory for organic acids production, particularly l-malic acid, through genetic manipulation. However, the traditional method of collecting A. spores inoculation labor-intensive and resource-consuming. In our study, we used CRISPR-Cas9 system to replace promoter brlA, a key gene in conidiation, with xylose-inducible xylP acid-producing strain RG0095, generating brlAxylP. When induced xylose submerged liquid culture, brlAxylP exhibited significant upregulation conidiation-related genes. This induction allowed us easily collect abundance (>7.1 × 106/mL) medium. Significantly, conidiation approach preserves substantial potential as foundational cellular platform biosynthesis acids, including but not limited acid. summary, study offers simplified strategy streamline preparation stage reduce labor material costs industrial acid production using species.

Fusarium infection has become a significant and widespread concern in modern agriculture, posing substantial threat to numerous essential crop plants. The pernicious fungal pathogen been causing destruction diverse areas, resulting severe consequences for both food security economic stability. Mycotoxins, which are secondary metabolites produced by wide variety of fungi, present serious risks the health humans animals have potential result considerable financial losses agricultural industries. genus is particularly infamous its capacity produce range mycotoxins. summarized study highlights recent progression fusarium towards various pivotal plants with their life cycle, morphological microscopic characteristics, pathogenicity factors. species well-known ability mycotoxins, such as zearalenones, fumonisins, Trichothecenes, Deoxynivalenol, Nivalenol, T-2 toxins detrimental that contaminate products, particular emphasis on grains cereals. new emerging mycotoxins producing challenges control human health. identification implementation efficient biocontrol strategies detoxify through Trichoderma, Plant Secondary Metabolites, Lactic Acid Bacteria, Edible Mushrooms, Enzymes measures guaranteeing safety protecting public mentioned approaches also discussed addressed. As an important intermediate in the tricarboxylic acid (TCA) cycle, l-malic (l-MA) is also one of 12 platform bulk chemicals with high added value. Owing to its various applications food, pharmaceuticals, cosmetics and industry, global l-MA market size growing year by year. Over last few decades, increasing concerns regarding fossil fuels depletion excessive CO2 emissions have led commitment fostering a green economy sustainable development. Alternatively, microbial fermentation has gradually attracted more attention. Here, this review summarizes common biosynthesis pathways compares differences between different chassis microorganisms as well. Moreover, regulation strategies genetic metabolic engineering process boost production are summarized, research status from cheaper substrates discussed. Finally, direction further exploration industrialized proposed, which provides theoretical guidance on promoting technological innovation industrial production.
