This study investigated the nitrogen removal performance of a three-stage AO reactor for refractory TN and changes in microbial community structure within activated sludge system under varying sodium chloride concentration conditions. Under an influent 0 g/L with sufficient carbon source, rates Total Nitrogen (TN), Chemical Oxygen Demand (CODcr), Ammonium (NH4+-N) remained stable at 98%, 99.7%, 99.9%, respectively. When increased to 20 g/L, activity AOB was significantly inhibited, efficiency dropping 83%, 89%, 70%, respectively, NAR increasing 91.97%. Analytical results demonstrated that both ammonia-oxidizing bacteria (AOB) nitrite-oxidizing (NOB) exhibited inhibited metabolic activities, NOB experiencing earlier functional impairment. NaCl concentrations ≤ 10 conventional via nitrification–denitrification (ND) dominant. exceeded due accumulation NO2−-N, phyla Planctomycetota Proteobacteria maintained dominance community, while partial nitrification (PN) denitrification pathways gradually replaced ND. Extracellular polymeric substance (EPS) secretion emerged as primary defense mechanism against salinity stress. Experimental findings informed proposed strategies including phased acclimatization salt-tolerance enhancement, EPS production regulation, targeted enrichment consortia, which collectively improved by 18.7–22.3% levels g/L. provides theoretical foundations technical references process optimization hypersaline industrial wastewater treatment systems.
