Abstract Heart failure (HF) is characterized by severely reduced cardiac function and tissue remodeling, driven complex multicellular regulatory processes. Extensive studies have generated molecular profiles at both bulk single-cell levels; however, systematic integration that describes tissue-wide changes as a of cell type coordination remains challenging. This disconnect hampers our understanding the interactions driving heart failure, limiting ability to translate insights into actionable therapeutic strategies. Here, we integrated transcriptional from tissues HF control patients across 25 studies, covering 1,524 individuals seven cell-types, delineate consensus associated with remodeling. Our analyses revealed conserved cellular events involving fibrotic, metabolic, inflammatory, hypertrophic mechanisms, fibroblasts playing central role in predicting cardiomyocyte stress. Further analysis fibroblast populations suggested their activation represents broad phenotypic shift rather than solely accumulating distinct states. The data within collection indicated responses types occur independently composition. Mapping independent programs demonstrated recovery after left ventricular assist device implantation aligns recovery, highlighting clinical relevance state. Background/Objectives: Determining appropriate cellular objectives is crucial for the system-scale modeling of biological networks metabolic engineering, reprogramming, and drug discovery applications. The mathematical representation can describe how cells manage limited resources to achieve goals within mechanistic environmental constraints. While rapidly proliferating like tumors are often assumed prioritize biomass production, mammalian cell types exhibit beyond growth, such as supporting tissue functions, developmental processes, redox homeostasis. Methods: This review addresses challenge determining trade-offs from multiomics data. Results: Recent advances in single-cell omics, modeling, machine/deep learning methods have enabled inference at both transcriptomic levels, bridging gene expression patterns with phenotypes. Altered body condition and diminished growth in wildlife the Alberta Oil Sands Region (AOSR) are prompting investigations into impact of oil sands industrial activity on region. Chemical constituents from bitumen-influenced waters, including process-affected water (OSPW), can disrupt endocrine signaling, leading to aberrant lipid accumulation altered glycemic control mammals. This study aimed investigate effects naphthenic acid fraction components (NAFCs), derived OSPW, energy homeostasis using McA-RH7777 rat hepatocyte model. Cells were exposed NAFCs at nominal concentrations 0, 0.73, 14.7, 73.4 mg/L for 24 48 h. We assessed gene expression related glucose metabolism measured triglyceride accumulation, glucose, fatty uptake. NAFC exposure (14.7 mg/L) reduced levels uptake increased beta-oxidation genes, suggesting a metabolic switch oxidation. substrate availability signifies shift cellular dynamics, potentially linked mitochondrial function. To this, we conducted adenosine triphosphate (ATP), membrane potential, terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) assays measure ATP levels, apoptosis, respectively. At both time points, resulted induced hyperpolarization, apoptosis. These results suggest that efficiency is compromised, necessitating adaptations maintain homeostasis. Given cells exhibit flexibility allows them dynamically respond changes availability, further demonstrated kynurenine-tryptophan ratio (KTR) serves as marker under these stress conditions. work provides mechanistic framework understanding how bitumen-derived organic contaminants may function living AOSR. findings support use molecular markers like KTR evaluate sub-lethal environmental health monitoring.
