ABSTRACT Chronic migraine (CM) is a disabling neurological disease. Astragaloside IV (AS‐IV), natural product derived from Astragalus membranaceus , shows great potential in treating chronic pain by relieving inflammation and oxidative stress. This study aimed to investigate the effects mechanisms of action AS‐IV on CM. An inflammatory soup comprising histamine, bradykinin, serotonin, prostaglandin E2 was used establish CM rat model, while lipopolysaccharide applied induce an response primary astrocytes. Pain threshold measurements were evaluate nociceptive hypersensitivity, qPCR Western blotting detect indicators synaptic protein expression, Golgi‐Cox staining observe dendritic spine density, transmission electron microscopy ultrastructure. Mitochondrial function stress assessed using JC‐1 staining, Mitotracker reactive oxygen species (ROS) quantification, glutathione content. pretreatment alleviated central sensitization ameliorated astrocyte activation neuroinflammation. mitochondrial dysfunction vitro, reduced nuclear translocation NF‐κB production IL‐1β, which reversed ROS scavengers vitro or respiratory chain disruptors vivo. Our indicates that can inhibit neuroinflammation alleviating mitigate CM, thereby providing experimental basis for A . prevention treatment.
