BRD4, a member of the BET family proteins, is extensively studied in cancer and known for its complex network within tumor cells, interacting with various transcription factors epigenetic regulatory proteins. However, impact BRD4 associated genes on hepatocellular carcinoma (HCC) remains unclear. HCC one leading causes cancer-related deaths globally, often diagnosed at an advanced stage, limiting treatment options. In this study, we curated BRD4-interacting and, through analysis TCGA-LIHC dataset, developed four-gene prognostic model comprising EZH2, KIF20A, G6PD, KIF2C. This demonstrated strong predictive power both training validation cohorts, high gene expression levels significantly correlating poor prognosis. Furthermore, our revealed significant immunological differences between risk groups identified increased drug sensitivity high-risk patients to certain medications, while decreased others. Experimental further indicated that combination inhibitor ZBC260 EZH2 CPI-169 synergistically enhanced apoptosis HCC. Collectively, study provides scientific rationale early diagnosis personalized therapy, offering new insights into resistance treatment.
