Comprehensive Summary Expressed protein ligation (EPL) provides a powerful tool to access large‐size proteins with precise structures. Existing methods for constructing the critical thioester EPL have predominantly relied on recombinant intein fusion expressed in Escherichia coli (E. ). Total Chemical Synthesis of Glucagon Glycoforms Reveals the Remarkable Influence of Natural Glycosylation DOI

Glycosylation plays a critical role in modulating protein and peptide properties, yet the impact of recently discovered natural mucin-type O-glycosylation on therapeutic peptides like glucagon remains underexplored due to challenges obtaining homogeneous research samples. Here, we address this challenge by developing streamlined multistep synthesis-one purification protocol, enabling production 21 glycoforms with systematically varied glycosylation patterns. Investigation these revealed remarkable two properties for glucagon: solubility increased over 870-fold, fibrillation was completely inhibited, even under stringent conditions, while biological activity elevating blood glucose levels preserved. Computational analyses indicated that sialylated glycans disrupt intermolecular associations solid state, enhancing solubility, shift secondary structures from β-sheets α-helices, mitigating fibrillation.
