Congenital hypothyroidism (CH) is a major health issue that can lead to intellectual disability if not detected and treated earlier. The preliminary screening program for neonatal CH in Indonesia gave provisional incidence of 1:2513. Newborn using dried blood spot sample the standard method detection, but it has limitations. Despite proven benefits screening, still faces significant challenges implementing nationwide program. This study aimed develop more sensitive accessible by creating monoclonal antibodies (mAbs) against thyroid-stimulating hormone (TSH).TSH protein was isolated from newborn cord confirmed Western blot analysis. Mice were immunized with purified TSH, hybridoma cell lines generated through fusion. Hybridoma supernatants screened TSH-specific ELISA. mAb highest titer dialysis. analysis presence TSH fraction at 28 kDa. Immunized mice showed increase antibody compared control group. clones secreting high-titer identified. research successfully produced mAbs it. They enable development rapid, point-of-care diagnostic tests, such as lateral flow immunoassays, which provide results within minutes. It will lay groundwork innovative tools significantly improve early diagnosis treatment this condition, particularly resource-limited settings.

Since the outbreak of new coronavirus, point-of-care diagnostics based on nucleic acid testing have become a requirement for development pathogen diagnostics, which require ability to accurately, rapidly, and conveniently detect pathogens. Conventional amplification techniques no longer meet requirements detection in low-resource, low-skill environments because they specialist equipment, complex operations, long times. Therefore, recombinant polymerase (RPA) is becoming an increasingly important method today’s technology it can amplify acids 20–30 minutes at constant temperature, greatly reducing dependence equipment technicians. RPA products are primarily detected through methods such as real-time fluorescence, gel electrophoresis, lateral flow assays (LFAs), other techniques. Among these, LFAs allow rapid within visualization results, offering convenient operation low cost. combination with LFA has significant advantages holds broad application prospects (POC) particularly low-resource settings. Here, we focus principles combined LFAs, their diagnosis, main limitations, some improvements methods.
