Long-term continuous imaging of endogenous HClO burst is great importance for the elucidation various physiological or pathological processes. However, most currently reported probes have failed to achieve this goal due their insufficient photobleaching resistance under a laser source. Herein, highly stable ratiometric probe, HFTC-HClO 1, which capable continuously monitoring over long period time, has been judiciously developed. Briefly, de novo development 1 mainly involved three main steps: (1) novel coumarins (HFTC 1–5) were designed and synthesized; (2) scaffold, HFTC 3, was selected through dye screening cell validation; (3) based on candidate constructed, finally its superior sensing properties toward HClO. Furthermore, can quantitatively measure levels in real samples with excellent recovery (>90.4%), use 1-coated test strips qualitative analysis also achieved. In addition, application long-term intracellular successfully demonstrated. Significantly, able visualize generated rheumatoid arthritis mouse model.

ABSTRACT Hypochlorous acid (HClO) is released by immune cells in the system, and it helps body fight off infections inflammation killing bacteria, viruses, other pathogens. However, tissue damage or apoptosis may also be induced excess HClO. On this basis, we designed probe TPE‐NS choosing tetraphenylethylene (TPE) as luminescent unit dimethylthiocarbamoyl chloride recognition site. By Gaussian's transition state calculations, HClO will cut photoinduced electron transfer (PET) effect of hydrolysis reaction, thus emitting strong fluorescence. has rapid excellent specificity for HClO, limit detection 7.27 μM. Finally, was successfully used visualization endogenous exogenous cell experiments.
