Abstract The development of redox‐sensitive molecular fluorescent probes for the detection redox changes in Plasmodium falciparum ‐parasitized red blood cells remains interest due to limitations current genetically encoded biosensors. This study describes design, screening and synthesis new pro‐fluorophores based on flavylium azido dyes coupled by CuAAC click chemistry alkynyl analogues plasmodione oxide, key metabolite potent redox‐active antimalarial plasmodione. photophysical electrochemical properties these were evaluated, focusing their fluorogenic responses. influence both status quinone length PEG chain separating fluorophore from electrophore was investigated. fluorescence quenching photoinduced electron transfer is reversible high amplitude oxidized forms reinstated reduced hydroquinone forms. Our results demonstrate that shortening has effect enhancing response, likely non‐covalent interactions between two chromophores. Superoxide anion (O2·–) and peroxynitrite (ONOO–), two important oxidants under oxidative stress, coexist in complex cell organism systems, playing crucial roles various physiological pathological processes, particularly neurodegenerative diseases. Ferroptosis is a type of cell death triggered by the iron-dependent accumulation lipid peroxides in cells. Diabetes, chronic metabolic disorder characterized hyperglycemia, can lead to various health complications. The process ferroptosis and progression diabetes are closely linked redox homeostasis, which regulated levels reactive oxygen sulfur species. Currently, there no fluorescent probes available monitor changes homeostasis during diabetes. Here, we report first endeavor create reversible near-infrared fluorogenic (NIRF) probe for monitoring reversal precise diagnosis. In vitro data demonstrated that NIR-CSTe could cyclically reversibly detect ONOO- GSH up four times with minimal loss fluorescence intensity. With help NIR-CSTe, observed HT-1080 cells, induced undergo erastin after being washed PBS 24 h then treated ferrostatin-1, showed recovery intracellular levels. contrast, treatment deferoxamine did not yield similar results. Lastly, was also utilized early diagnosis efficacy assessment relation ONOO-/GSH balance, results illustrating combined administration metformin empagliflozin more effective than using either drug alone.
