Label-free surface-enhanced Raman spectroscopy (SERS) combined with machine learning (ML) techniques presents a promising approach for rapid pathogen identification. Previous studies have demonstrated that purine degradation metabolites are the primary contributors to SERS spectra; however, generating these distinguishable spectra typically requires long incubation time (>10 h) at room temperature. Moreover, lack of attention spectral variations between strains same bacterial species has limited generalizability ML models in real-world applications. To address issues, we investigated temperature-induced alterations metabolism and found robust could be obtained within just 1 h by heating samples 60 °C. Our study further revealed pathogens exhibit multiple fingerprint patterns across strains, rather than uniform signature. enhance practicality, optimized training them on data sets capturing all relevant fingerprints validated separate strains. The SoftMax classifier achieved 100% accuracy identifying both laboratory clinical specimens 17 h. Additionally, platform over 91% distinguishing drug-resistant such as methicillin-resistant Staphylococcus aureus carbapenem-resistant Klebsiella pneumoniae, 99.66% differentiating specific species, enterohemorrhagic Escherichia coli. This accelerated, metabolism-based offers highly alternative diagnosis infections.
