ABSTRACT
The
accurate
and
sensitive
detection
of
low‐abundance
cancer‐related
biomarkers
in
blood
remains
a
key
technical
challenge
clinical
applications.
Herein,
simple
sandwich‐type
electrochemical
immunosensor
based
on
stable
hydrogen‐bonded
cobalt‐porphyrin
framework
(Co‐HOF)
was
successfully
developed
for
the
ultrasensitive
biomarker,
carcinoembryonic
antigen
(CEA).
antibody‐modified
Co‐HOF
forms
sandwich
structure
with
CEA
aptamer
electrode
exclusively
presence
CEA,
enabling
specific
CEA.
signal
increased
linearly
concentration
demonstrating
wide
linear
range
(0.001–50
ng
mL
−1
)
low
limit
(0.22
pg
),
surpassing
performance
commercial
ELISA
kits
most
reported
methods.
sensor
employed
spiked
human
serum,
recoveries
ranging
from
85.04%
to
105.20%.
Additionally,
we
collected
samples
colorectal
cancer
patients
healthy
individuals
clinically
validate
sensor,
observing
that
levels
progression.
results
showed
strong
consistency
(
R
2
=
0.995)
those
obtained
kits,
proposed
sensor's
practicality
related
biomarkers.
