Abstract Thermo‐responsive polymers have emerged as a cutting‐edge tool in nanomedicine, paving the way for innovative approaches to targeted drug delivery and advanced therapeutic strategies. These “smart” respond temperature changes, enabling controlled release pathological environments characterized by high temperatures. By exploiting their unique phase transition, occurring at lower or upper critical solution temperatures (LCST UCST), these systems ensure localized action, minimizing collateral damage healthy tissues. The integration of into nanoparticles with hydrophilic shells hydrophobic cores enhances stability biocompatibility. Furthermore, polymer engineering allows precise modulation LCST UCST through adjustments composition hydrophilic‐lipophilic balance, optimizing responsiveness specific applications. In addition delivery, thermo‐responsive are gaining attention several fields such gene therapy imaging. Therefore, this review explores chemical structural diversity nanoparticles, emphasizing ability encapsulate drugs effectively. Second, highlights potential redefine treatment paradigms, providing comprehensive understanding mechanisms, applications, future perspectives biomedical research.
