Significance As a critical component of healthy memory management system, forgetting has received increasing attention. Studies across multiple species support important roles actin remodeling in forgetting. However, the underlying molecular mechanisms remain unclear. In Drosophila , Rac1 and Cdc42, 2 Rho GTPases that act as signaling hubs to coordinate remodeling, were reported mediate labile consolidated memories, respectively. Here, we showed Cdc42 exert their effects on by acting through different polymerization pathways, Rac1/SCAR/Dia Cdc42/WASp/Arp2/3 complexes. Memory consolidation theory suggests that once memory formation has been completed, is maintained at a stable strength and incapable of further enhancement. However, the current study reveals even long after formation, contextual fear could be enhanced. Such unexpected enhancement possible because dynamically an intermediate level allows for bidirectional regulation. Here we find both Rac1 activation expression α2-chimaerin are stimulated by single-trial conditioning. sustained activity mediates reversible forgetting, acts as molecule reverses forgetting to sustain through inhibition during maintenance stage.
