Autism is a class of developmental disorders with extremely high rates disability, affecting patients throughout their lives. There no cure to date clinically, and early rehabilitation interventions can improve some the behavioral problems autistic patients, but these are limited by age often have minimal effects in older adults autism. Early diagnosis also necessary while developing effective autism therapies. At present, dependent on search for markers an attempt screen differentially expressed proteins using high-throughput assays, such as synaptic scaffolding proteins, microtubule-associated apolipoproteins, immunoglobulin G complement factor-related etc. It would be big step forward mechanistic studies if valid biomarker could found.

Consequently, understanding neural circuit mechanisms underlying has become a major focus in neuroscience. behaviors, which encompass wide range expressions phases, regulated by complex networks. In this review, we summarize recent progress identifying circuits involved different types general investigation, preference, mating, aggression, parenting, prosocial dominance behaviors. We also outline associated with deficits ASD, schizophrenia, PTSD. Given pivotal role rodents behavior research, our review primarily focuses on these animals. Finally, propose future research directions, development specific behavioral paradigms, identification motor output, integration activity, transcriptome, connectome data, multifunctional roles neurons multiple targets, among brain regions.

Abstract Dopamine (DA) neurons in the ventral tegmental area (VTA) promote social brain functions by releasing DA onto nucleus accumbens neurons, but it remains unclear how VTA communicate with cortical neurons. Here, we report that medial prefrontal cortex (mPFC)-lateral hypothalamus (LH)-VTA pathway contributes to deficits mice IRSp53 deletion restricted excitatory ( Emx1-Cre;Irsp53 fl/fl mice). LH-projecting mutant mPFC display abnormally increased excitability involving decreased potassium channel gene expression, leading excessive synaptic input LH-GABA A circuit-specific also increases neuronal and induces deficits. show a compensatory decrease excitability, weakening inhibitory LH GABA -VTA subsequently over-activating VTA-GABA over-inhibiting VTA-DA Accordingly, optogenetic activation of improves mice. Therefore, mPFC-LH

Abstract Schizophrenia (SCZ) is a severe psychiatric and neurodevelopmental disorder. The pathological process of SCZ starts early during development, way before the first onset psychotic symptoms. DNA methylation plays an important role in regulating gene expression dysregulated involved pathogenesis various diseases. methylated immunoprecipitation‐chip (MeDIP‐chip) performed to investigate genome‐wide dysregulation peripheral blood mononuclear cells (PBMCs) patients with first‐episode (FES). Results show that SHANK3 promoter hypermethylated, this hypermethylation (HyperM) negatively correlated cortical surface area left inferior temporal cortex positively negative symptom subscores FES. transcription factor YBX1 further found bind HyperM region induced pluripotent stem (iPSCs)‐derived interneurons (cINs) but not glutamatergic neurons. Furthermore, direct positive regulatory effect on confirmed cINs using shRNAs. In summary, suggests potential neuropathological mechanism underlying SCZ. results also suggest PBMCs can serve as biomarker

Accumulated studies have determined the changes in functional connectivity autism spectrum disorder (ASD) and spurred application of machine learning for classifying ASD. Graph Neural Network provides a new method network analysis brain disorders to identify underlying features associated with deficits. Here, we proposed an improved model Isomorphism (GIN) that implements Weisfeiler-Lehman (WL) graph isomorphism test learn while taking into account importance each node classification improve interpretability algorithm. We applied on multisite datasets resting-state connectome from Autism Brain Imaging Data Exchange (ABIDE) after stringent quality control. The outperformed other commonly used methods five different evaluation metrics. also identified salient ROIs visual frontoparietal control networks, which could provide potential neuroimaging biomarkers ASD identification.

Abstract Traumatic brain injury (TBI) is often more complicated than a single head injury. An extreme example of this point may be military service members who experience spectrum exposures over prolonged period under stressful conditions. Understanding the effects complex can inform evaluation and care to prevent persistent symptoms. We designed longitudinal series non‐invasive procedures in adult mice evaluate mild stress exposures. assessed anxiety, depression, sleep–wake dysfunction as symptoms that impact long‐term outcomes after TBI. Unpredictable chronic (UCMS) was generated from varied sequence environmental stressors distributed within each 21 days. Subsequently, received blast combined with closed‐head TBI on 5 days at 24‐h intervals. In males females, UCMS induced anxiety without depressive behavior. A major finding reproducible through 6‐ 12‐month time points male repetitive plus events, or surprisingly just alone. Specifically, exhibited hypersomnia increased sleep during active/dark phase fragmentation longer wake bouts. Sleep–wake not found events alone, females any These results identify sex differences important considerations for dysfunction. Furthermore, impaired wakefulness similar excessive daytime sleepiness reported patients, including patients TBI, which warrants further clinical screening, care, treatment development.

Abstract Prefrontal cortical maturation coincides with adolescent transitions in social engagement, suggesting that it influences development. The anterior cingulate cortex (ACC) is important for interaction, including ACC outputs to the basolateral amygdala (BLA). However, little known about ACC-BLA sensitivity environment and if this changes during maturation. Here, we used brief (2-hour) isolation test immediate impact of changing on circuit subsequent shifts behavior adult rats. We found optogenetic inhibition reduced isolation-driven facilitation interaction across ages. Isolation increased activity neurons ages, but altered influence BLA an age-dependent manner. inhibitory stimulation a frequency-dependent manner adults, uniformly suppressed ACC-driven adolescents. This work identifies alterations circuit, itself as essential region sensitive regulates its both adults

Abstract The purpose of this study was to determine the effect Cz high‐definition 5‐channel tDCS (HD‐tDCS) on social function in 4–12 years‐old children with autism spectrum disorder (ASD). This a randomized, double‐blind, pseudo‐controlled trial which 45 ASD were recruited and divided into three groups sex, age, rehabilitation treatment as control variables. Each group 15 randomly administered active HD‐tDCS central anode, left dorsolateral prefrontal cortex (F3) sham anode 14 daily sessions 3 weeks. Social Responsiveness Scale Chinese Version (SRS‐Chinese Version) compared 1 week after stimulation values recorded prior stimulation. At end treatment, both anodal DLFPC decreased measures SRS‐Chinese Version. total score by 13.08%, cognition 18.33%, communication 10.79%, significantly improved over group, especially young middle low function. There no significant change subscale group. In F3 13%, autistic behavior 19.39%, 14.39%, all improved. However, there difference between DLPFC conditions. may be an effective for dysfunction ASD.
