Abstract Background Synaptotagmin 11 (SYT11) plays a pivotal role in neuronal vesicular trafficking and exocytosis. However, no independent prognostic studies have focused on various cancers. In this study, we aimed to summarize the clinical significance molecular landscape of SYT11 tumor types. Methods Using several available public databases, investigated abnormal expression different types its potential association with prognosis, methylation profiling, immune infiltration, gene enrichment analysis, protein–protein interaction identified common pathways. Results TCGA Genotype-Tissue Expression (GTEx) showed that was widely expressed across corresponding normal tissues. Survival analysis correlated prognosis seven cancer Additionally, mRNA not affected by promoter methylation, but regulated certain miRNAs associated patient prognosis. vitro experiments further verified negative correlation between miR-19a-3p human colorectal, lung, renal cell lines. Abstract GABA B receptors (GBRs), the G protein-coupled for GABA, regulate synaptic transmission throughout brain. A main function of GBRs is gating Cav2.2-type Ca 2+ channels. However, cellular compartment where stable GBR/Cav2.2 signaling complexes form remains unknown. In this study, we demonstrate that vesicular protein synaptotagmin-11 (Syt11) binds to both auxiliary GBR subunit KCTD16 and Cav2.2 Through these dual interactions, Syt11 recruits channels post-Golgi vesicles, thus facilitating assembly complexes. addition, stabilizes at neuronal plasma membrane by inhibiting constitutive internalization. Neurons knockout mice exhibit deficits in presynaptic channels, reduced neurotransmitter release, decreased GBR-mediated inhibition, highlighting critical role expression These findings support acts as a scaffold protein, aiding from low-abundance components within transport vesicles. This mechanism enables insertion pre-assembled functional units into membrane.
