This review discusses the chemical properties, synthesis and detection, biological functions of a molecular group cis-2-unsaturated fatty acids, containing acid carbon chains various lengths cis double-bond configurations, known as diffusible signaling factor family (DSFF). Early postulation conserved nature DSFF among Gram-negative bacteria have now been challenged by latest evidences that unraveled their presence in other distinct microorganisms. Over last decade, significant depth breadth understanding has made on multifaceted DSFFs bacteria, interactions with evolutionarily divergent fungi, plants insects small animals. While knowledge within microbial systems is still developing, molecules such BDSF, DSF, SDSF found to modulate virulence, cell adhesion, biofilm formation dispersion, motility, antimicrobial tolerance. Given capacity influence ecosystems rapid emergence novel DSFF-like molecules, it critical identify full spectrum members better understand this complex messenger system they offer potential be exploited development new therapeutic strategies combat rising global healthcare threat resistance. narrative therefore provides broad picture quorum sensing core foundation built from seminal literature while highlighting developments field.

SUMMARY Diffusible signal factor (DSF)-family quorum sensing (QS) signals are widely utilized by many bacterial pathogens to modulate various biological functions and virulence. Previous studies showed that cis -2-decenoic acid ( -DA) is involved in modulation of biofilm dispersion Pseudomonas aeruginosa , but its signaling mechanism remains vague. Here, we report -DA regulates the physiology virulence P. through long-chain fatty acid-CoA ligase FadD1. Cis specifically binds FadD1 with high affinity enhances binding promoter DNA region lasR . Further analysis a response regulator DNA-binding leucine zipper motif control transcription target genes. Moreover, exhibited catalytic activity on -2-dodecenoic (BDSF) Burkholderia cenocepacia enhanced competitiveness Together, our work presents new DSF-type QS system, which highlighted receptor evolved from canonical enzyme metabolism. Abstract Figure Highlights The acts as global transcriptional at top hierarchy homolog fluorescens could bind gene responding -DA. In brief human pathogen antibiotic resistance wide range dynamic defenses makes it an extremely challenging organism treat modern-day medicine. It employs regulate pathogenesis. Song et al. demonstrate controls important novel signal.
