Humans are inevitably exposed to micro- and nanoplastics (MP/NP). These particles able cross the biological barriers enter bloodstream with levels close 1.6 µg mL-1; MP/NP have been detected in placentas meconium of newborns. However, consequences this exposure on integrity, development functions human placenta not documented. In study, trophoblasts purified from at term were for 48 h, two different sizes polystyrene nanoparticles (PS-NP) 20 nm (PS-NP20) 100 (PS-NP100), environmental supra-environmental concentrations (0.01-100 mL-1). Cell viability, oxidative stress, mitochondrial dynamics, lysosomal degradation processes, autophagy, inflammation/oxidative responses placental endocrine angiogenic assessed. PS-NP size determines their internalization rate behavior trophoblasts. Indeed, PS-NP20 more rapidly translocated, accumulated lysosomes as shown by confocal TEM imaging. They induce higher cytotoxicity than PS-NP100, early 1 mL-1 (p < 0.05). addition, they a pro-inflammatory cytokines response: IL-1ß is induced 0.01 both nanoparticle sizes; IL-6, TNF-α overexpressed only For first time, we report that disrupt function, observed decreased hCG release found blood. This work, provides an in-depth vitro assessment effects placenta.

Nanoplastics (NPs) represent an escalating hazard to both humans and the ecosystem due their pervasive presence. This review delves into (i) widespread occurrence of NPs across different environmental matrices, including food; (ii) routes estimates for human exposure; (iii) mechanisms blood–brain barrier (BBB) crossing; (iv) implications health, with a specific focus on molecular features associated neurotoxicity neurodegenerative processes. The impact central nervous system, ability cross BBB underpinning mechanisms, potential initiate by fostering β-amyloid aggregation, interactions metallo-enzymes (such as superoxide dismutase) are elucidated. analysis transcriptomics epigenomic results, microRNA dysregulation, unveil how could contribute neurological disorders. need considering overlaps among diverse pathogenetic when probing effects is discussed. Additional urgent needs development reliable in vitro models studies able mimic complexity system exposure such more environmentally relevant NPs. Finally, extremely sensitive detection methodologies quantify biological matrices pressing priority.

Water-soluble polymers (WSPs) are a particular category of that, due to their capability be soluble in water, come out the classic definition plastic and therefore also from its regulation control, representing possible new environmental problem considering number consumer products which they contained. For this reason, aim study was evaluate adverse effects three most used WSPs (polyacrylic acid - PAA, polyethylene glycol PEG, polyvinylpyrrolidone PVP), administered at relevant concentrations (0.001, 0.5 1 mg/L) Danio rerio (zebrafish) embryos up 120 h post fertilization. To assess WSP toxicity molecular, cellular organism level we an integrated ecotoxicological approach both biomarkers high-throughput technology based on gel-free proteomics. The main results showed how all up-regulated many proteins (up 74 specimens exposed mg/L PVP) with wide range molecular functions involved numerous pathways specimens. On other hand, measurement PAA PVP were able activate antioxidant machinery following over-production reactive oxygen species, while PEG produced no significant changes measured. Based obtained results, use application should revised regulated.

Nanoplastics (NPs), as a new type of pollutant with size small than 1 μm, are ubiquitous and harmful to organisms. There has been an increasing amount research concerning the effects NPs on organisms over recent years, especially aquatic animals. However, there is limited study impact mollusk cephalopods. In this research, Sepia esculenta , belonging Cephalopoda, Coleoidea, Sepioidea, was selected explore caused by exposure. The S. larvae were exposed polystyrene (PS-NPs) diameter 50 nm (100 mg/L) for 4 h. detection oxidative stress biomarkers displayed obvious increase in SOD (superoxide dismutase) activity MDA (malondialdehyde) level. Then, RNA-Seq performed response at mRNA transcriptome analysis demonstrated that expression 2,570 genes affected PS-NPs. Besides, signaling pathways ribosome, ribosome biogenesis eukaryotes, proteasome, MAPK enriched. This not only provides novel references understanding mechanisms induced NPs, but also reminds us follow interest influence acute exposure NPs.

Microplastic pollution represents an emerging problem of great interest in the public domain last decade; addition, it overlaps with another delicate - pharmaceutical products that can have negative effects on environment and people, even small amounts. The main purpose this study was to assess biochemical behavioral exposure adult zebrafish (Danio rerio) polyethylene (PE), polypropylene (PP) valproic acid (VPA), respectively their mixtures possible situations natural aquatic environments. In terms responses, sociability appears be more impaired PP group after 5 days exposure. mechanisms affected are those swimming performance than sociability. Even more, VPA increases presence arm conspecifics but decreases mobility locomotion, indicating a anxiety mechanism. decrease aggressiveness, especially case PE+VPA group, where reaches super low level compared control, which could endanger species nature. Regarding anxiogenic effect, PE act differently, if has opposite side is shows bolder agitated behavior. All four variants showed changes indicative toxicity from first dose.
