The introduction of antibiotic-resistant bacteria into riverine systems through the discharge wastewater treatment plant (WWTP) effluent and agricultural waste poses significant health risks. Even when not pathogenic, these can act as reservoirs for antibiotic resistance genes (ARGs), transferring them to pathogens that infect humans animals. In this study, we used fluorescence in situ hybridization, qPCR, metagenomics investigate how anthropogenic activities affect microbial abundance resistome along Holtemme River, a small river Germany, from near-pristine human-impacted sites. Wastewater treatment plants (WWTPs) are thought to be a major disseminating source of antibiotic resistance (AR) the environment, establishing crucial connection between human and environmental resistome. The objectives this study were determine how wastewater effluents impact microbiome resistome freshwater fish, identify potential AR-carrying clinically relevant pathogens in these matrices. We analyzed influent effluent from four WWTPs three metropolitan areas Ohio, USA via shotgun metagenomic sequencing. also sequenced river water fish guts reaches (upstream, at WWTP outfall, downstream). Notably, we observed decline diversity AR gene abundance receiving river. found significant differences by reach trophic level (diet) beta-diversity gut microbiomes. SourceTracker revealed that 0.443 0.248 more was sourced outfall downstream locations, respectively, compared upstream fish. Additionally, bacteria public health concern annotated samples, indicating for exposure. In summary, our findings show continued role as reservoir underscores considerable discharge on aquatic wildlife, which highlights One Health nature issue.

The aquatic environment plays a central role in the selection and spread of antimicrobial resistance genes (ARGs). Using metagenomic approaches, several studies have provided comprehensive view resistome across different ecosystems. However, unique systems, such as oceanic blue holes, remain largely unexplored. In this study, free-living particle-associated shotgun metagenome sequencing data, collected Sansha Yongle Blue Hole (South China Sea) water column, were analyzed to reveal (total content ARGs within defined bacterial community) potential for horizontal gene transfer detected ARGs. diversity overall significantly decreased with depth, although trend was driven by community, while it remained stable communities. Additionally, latter showed higher frequency co-occurring mobile genetic elements on same contigs, frequent plasmid localization these genes. Overall, bacteria proved essential sustaining diversity. Furthermore, findings suggest potentially rate ARG microbial communities along column ensuring persistence resistances deep marine waters.

Abstract Background Antimicrobial resistance is a major global health concern, with the environment playing key role in its emergence and spread. Understanding relationships between environmental factors, microbial communities, mechanisms vital for elucidating resistome dynamics. In this study, we characterized of Baltic Sea evaluated how gradients spatial variability, alongside communities associated functional genes, influence diversity composition across geographic regions. Results We analyzed metagenomes benthic sediments from 59 monitoring stations 1,150 km distance Sea, revealing an comprised predicted antimicrobial genes (ARGs) against 26 antibiotic classes. observed variation profile, higher northern regions decline dead zones southern areas. The combined effects salinity temperature gradients, nutrient availability, created complex landscape that shaped distribution ARGs. Salinity predominantly influenced ARG composition, leading to clear distinctions high-saline those lower mid-level salinity. Furthermore, our analysis suggests community mobile genetic elements might be crucial shaping composition. Conclusions presented were identified as primary factors influencing regions, availability further these patterns Sea. Our study also highlighted interplay resistance, ecosystem, underscoring potential element distribution. modulate resistomes will help predict impact future changes on aquatic ecosystems.

However, distribution and influencing factors antibiotic resistance genes (ARGs) this lake are still controversial. Methods To solve problem, we used metagenomic sequencing investigate in-fluencing ARGs CL. Results Our findings revealed existence nine kinds ARGs, including 45 specific genes. The abundant types were multidrug, bacitracin, polymyxin, macrolide lincosamide streptogramin, aminoglycoside. Multiple microorganisms undeniable ARG reservoirs, although they not dominant species microbiota. results also showed that both microbiota physiochemical played important roles shaping distributions Specifically, levels PO4-P (0.5927) total phosphorus (0.4971) had a greater impact than nitrogen (0.0515), NO 3- N (0.0352), NO2-N (−0.1975), NH3-N (−0.0952). Conclusions These provide valuable insights into lakes.
