The gut microbiome is altered after bariatric surgery and associated with weight loss. However, the commensal bacteria involved underlying mechanism remain to be determined. We performed shotgun metagenomic sequencing in obese subjects before longitudinally sleeve gastrectomy (SG), found a significant enrichment microbial species Clostridia bile acid metabolizing genes SG treatment. Bile profiling further revealed decreased primary acids (PBAs) increased conjugated secondary (C-SBAs) SG. Specifically, glycodeoxycholic (GDCA) taurodeoxycholic (TDCA) were at different follow-ups SG, abundance of body reduction. Obesity is increasingly becoming a challenge with China's economic development. There an urgent need to identify more affordable methods combat this condition. Paederia scandens (PS), cost-effective herbal remedy widely used in China for treating inflammation and pain, shows potential regard. To investigate its anti-obesity mechanisms, we established high-fat diet (HFD)-induced obesity model mice. The obese mice subsequently received daily oral gavage of PS extract 21 consecutive days. Upon the completion experiment, blood samples were collected analyze lipid profiles, including total cholesterol (TC), triglycerides (TG), high-density lipoprotein (HDL-C), low-density (LDL-C). Abdominal adipose tissue was subjected hematoxylin-eosin (HE) staining histological analysis, while fecal underwent 16S rRNA sequencing assess gut microbiota composition. Our findings revealed that supplementation significantly reduced body weight, metabolism biomarkers, adipocyte size. treatment also restored microbial diversity, 19 specific taxa 25 KEGG pathways identified as mediators effects. Serum metabolomics analysis implicated L-ascorbic acid, stevioside, allopurinol, gingerol, along amino acid energy pathways, mechanism PS. Microplastics (MPs) and antibiotic tetracycline (TC) are widespread in the environment constitute emerging combined contaminants. Young individuals particularly vulnerable to agents that disrupt intestinal health development. However, effects of MPs TC remain poorly understood. In this study, we developed a young mouse model exposed polystyrene MPs, either alone or combination with for 8 weeks simulate real-life dietary exposure during early life. Our findings revealed concurrent caused most severe barrier dysfunction driven by inflammatory activation oxidative imbalance. Moreover, reduced abundance potential probiotics while promoting growth opportunistic pathogens. Metagenomic analysis further indicated co-exposure enhanced bacteria carrying resistance genes (ARGs) virulence factor (VFGs), contributing dissemination potentially harmful genes.
