Preparation of chiral metal organic framework modified silica monolithic capillary column and its application in enantioseparations of chiral drugs DOI

Metal organic frameworks (MOFs) are crystalline compounds composed of metal ions (or clusters) and ligands. Chiral MOFs have been successfully utilized as novel materials for the separation chiral enantiomers by chromatography, demonstrating excellent performance. In this study, a MOF-modified silica monolithic capillary column was used pressurized electrochromatography. First, MOF (Co-glycyl-L-glutamic acid, Co-L-GG) synthesized. This then to prepare via one-step in situ polymerization method. The optimal conditions preparing were determined follows: Co-L-GG amount, 5 mg; polyethylene glycol 0.96 tetramethoxysilane dosage, 3.6 mL; trimethoxymethylsilane 0.4 mL. Next, effects on drugs investigated. Under an applied voltage -20 kV mobile phase consisting acetonitrile 20 mmol/L disodium hydrogen phosphate (80∶20, v/v), six separated within 3 min, with baseline achieved amlodipine, fluvastatin, tryptophan. Moreover, prepared exhibited good reproducibility stability. Finally, molecular docking studies conducted using AutoDock explore recognition mechanism, results analyzed Discovery Studio. indicated that larger differences binding free energy between correlated higher enantioselectivity factors. However, correlation did not necessarily lead increase resolution. Co-L-GG, which is enriched primary amines, secondary carbonyl groups, demonstrated enantiomeric capability. conclusion, study demonstrates can be effectively functional monomers columns, highlighting their significant potential analysis compounds. comprehensive exploration synthesis, characterization, applications these will help provide valuable insights into development advanced technologies.
