Rhopalosiphum padi is a global agricultural pest which had developed resistance to different insecticides. The ATP-binding cassette (ABC) transporter plays an important role in insecticide resistance. However, ABC transporters' and regulatory mechanism mediating R. 's response pyrethroids are unclear. In this study, we found that RpABCG23L was significantly overexpressed the pyrethroid-resistant strains of padi. Knockdown increased susceptibility lambda-cyhalothrin bifenthrin. Luciferase reporter gene analysis showed miR-275 binds coding region down-regulates its expression. Injection mimics reduced expression bifenthrin, while inhibitor injection enhanced tolerance both results provide theoretical basis for understanding miRNA-mediated pyrethroid resistance, open up new way development miRNA-based biopesticides.

Abstract The use of synthetic pesticides carries a significant risk pests developing resistance, leading to decreased pesticide effectiveness. ATP‐binding cassette (ABC) transporters, especially the ABCC subfamily members, have been suggested act as efflux pumps for various pesticides, thereby contributing resistance. So far, identification potential substrates insect ABC transporters is most often based on quantification transcript in arthropods. Here, we screened and identified ABCC‐9C from Tribolium castaneum an vitro ATPase activity assay. Together with affinity evaluation‐, cytotoxicity analysis‐, RNA interference‐based bioactivity tests, revealed that insecticides, carbofuran, buprofezin, are Tc ABCC‐9C. Additionally, amphipathic translocation channel transmembrane domain formed by 8 helices. Molecular docking both carbofuran buprofezin bind at same site within via hydrophobic interactions. These findings indicate might play critical role multi‐pesticide providing target managing resistance laying groundwork future pest control strategies. Given conservations among experimental model developed this study can be also applied identify other well predict insecticide mediated transporters.
