To determine the main components of Astragalus membranaceus (Fisch.) Bge. (A. membranaceus, Huang Qi), Astragaloside IV (AIV) and polysaccharides (AP), to characterize their properties, evaluate in vivo efficacy, analyze drug diffusion using dissolving microneedle (DMN) technology vivo. Respectively, AIV- AP-loaded DMNs comprising chitosan (CTS) polyvinyl alcohol (PVA) were prepared via dual-mold forming. Their morphology, mechanical solubility, skin irritation characteristics tested. In efficacy was assessed cyclophosphamide-induced immunosuppressed mice, AIV AP by conventional methods compared, rheological properties AIV-CTS-PVA AP-CTS-PVA mixtures measured. Subcutaneous dissolution absorption microneedles (MNs) at low doses (50% – 17% intraperitoneal injection 12% 4% intravenous injection) reduced spleen index acid phosphatase activity mouse models, increased thymus index, achieved equivalent or better systemic therapeutic effects. Compared with injections, controllable solid-liquid conversion through delivery CTS-PVA MNs, resulting highly localized aggregation within 48 h, reducing initial explosive effect drug, achieving stable slow release. The present study enhances our understanding remote effects drug-loaded from a traditional Chinese medicine (TCM) perspective, thereby promoting development precise efficient TCM further expanding drug-loading range application scenarios for DMNs.
