We analyzed data from the Global Burden Diseases EUROSTAT. There was a significant increase expenditure health 1980 2019. period 1980-2000, despite higher spending, increased in almost all European countries. During 2000-2019, Western Countries Poland, Estonia, Latvia, Slovenia there correlation between decrease ASMR. The most important 1980-2000 2000-2019 proportion devoted to preventive care use aspirin statins. Information about have leads decreased aneurysm related burden.

Circulating amino acid levels can be altered in arterial aneurysms and dissection, but the relationships between them is unclear. The present study investigated causal relationship circulating dissection via bidirectional two-sample Mendelian randomization (MR). A MR analysis was used. Forward performed with as exposure outcomes. Reverse exposures data were analyzed using five analytical methods: inverse-variance weighted (IVW), MR‒Egger, median, simple, methods. IVW used main method, other methods for supplementary analyses. Heterogeneity assessed Cochran's Q test, horizontal pleiotropy intercepts from MR‒Egger regression. genome-wide association (GWAS) acids obtained IEU open GWAS database Catalog database. Finngen consortium version R10. tyrosine level negatively correlated (P = 0.00211, OR: 0.57, 95 % CI: 0.40, 0.82). Aortic decreased glycine 0.00168, 0.98, 0.99). Through analysis, we found that aortic reduced glycine. Our findings support a possible interaction dissection.

Thoracic aortic aneurysms (TAAs) are a major cause of death owing to weaker blood vessel walls and higher rupture rates in affected individuals. Vascular smooth muscle cells (VSMCs) the predominant cell type within wall their dysregulation may contribute TAA progression. Enhancer zeste homolog 2 (EZH2), histone methyltransferase, is involved several pathological processes; however, biological functions mechanisms underlying VSMC phenotype transition vascular intimal hyperplasia remain unclear. The present study aimed determine involvement EZH2 mediating function development TAAs. expression was revealed be elevated patients with thoracic dissection mouse model through western blotting reverse transcription‑quantitative PCR experiments. Subsequently, established using β‑aminopropionitrile. In vitro, EdU labeling, Transwell assay, wound healing assay hematoxylin‑eosin staining that knocking down Ezh2 gene could reduce proliferation, invasion, migration, calcification primary cells. Flow cytometry analysis found deficiency increased apoptosis. Depletion VSMCs promoted transformation from synthetic contractile phenotype. Using RNA‑sequencing analysis, it demonstrated regulated group genes, including integrin β3 (Itgb3), which critically extracellular matrix signaling pathway. qChIP occupies Itgb3 promoter, thereby suppressing Itgb3. promotes invasion VSMCs, this promoting effect partially reversed by co‑knocking In conclusion, identified previously unrecognized EZH2‑ITGB3 regulatory axis thus provides novel mechanistic insights into pathophysiological EZH2. serve as potential target for management

In observational and experimental studies, diabetes has been reported as a protective factor for aortic dissection. 3-Hydroxybutyrate, key constituent of ketone bodies, found to favor improvements in cardiovascular disease. However, whether the effect on dissection is mediated by 3-hydroxybutyrate unclear. We aimed investigate causal effects risk mediating role them through two-step Mendelian randomization.

Aortic aneurysm and dissection (AAD) are severe vascular diseases with high mortality rates. However, the causal relationship between serum uric acid levels occurrence of AAD remains a subject controversy. To address this issue, we conducted two-sample Mendelian randomization (MR) analysis to investigate whether there is association these factors. We obtained single-nucleotide polymorphisms (SNPs) data related from FinnGen study on UK Biobank. Various MR methods, including inverse variance weighted (IVW) analysis, MR-Egger regression median contamination mixture method, were employed assess risk AAD. Sensitivity was evaluate stability reliability results. The findings revealed positive aortic (AA) (odds ratio [OR] = 1.200, 95 % confidence interval [CI]: 1.020–1.400, P 0.0239). no significant correlation observed (AD) (OR 0.893, CI 0.602–1.326, 0.576). Our study, which identified AA. did not observe AD.

However, the causality them mediators this association not been identified. This study employs bidirectional multivariable Mendelian randomization (MR) to investigate dissection identify relationship. Methods: We first investigated causal using two-sample MR, with inverse-variance-weighted method as principal analysis technique. Subsequently, we applied MR determine probable mediators. Results: Genetically levels were linked (odds ratio [OR]: 1.160, 95% confidence interval [95% CI]: 1.011–1.332, P = 0.034), high-density lipoprotein cholesterol (HDL-C) mediating relationship, accounting for 10.2% effect. No was found Conclusions: Serum is factor aneurysm, mediated through HDL-C. Monitoring HDL-C in patients hyperuricemia essential prevent slow progression aneurysm.

Aortic aneurysm/dissection (AAD) poses a life-threatening cardiovascular emergency with complex mechanisms and notably high mortality rate. Zebrafish (Danio rerio) serve as valuable models for AAD due to the conservation of their three-layered arterial structure genome that humans. However, existing studies have predominantly focused on larval zebrafish, leaving gap in our understanding adult zebrafish. In this study, we utilized β-Aminopropionic Nitrile (BAPN) impregnation induce both Following induction, zebrafish exhibited 28% widening dorsal aortic diameter (p < 0.0004, n = 10) arch malformations, malformation rate 75% (6/8). Conversely, showed 41.67% (5/12) 22 days post-induction. At time point, area had expanded by 2.46 times 0.009), vessel wall demonstrated significant thickening (8.22 ± 2.23 μM vs. 26.38 10.74 μM, p 0.05). Pathological analysis revealed disruptions smooth muscle layer, contributing 58.33% aneurysm Moreover, expression levels acta2, tagln, cnn1a, cnn1b were decreased, indicating weakened contractile phenotype. Transcriptome sequencing overlap between molecular features tissues post-BAPN treatment those patients. Our findings present straightforward practical method generating using BAPN.
