Abstract Parallel ON and OFF (positive- negative-contrast) pathways fundamental to vision arise at the complex synapse of cone photoreceptors. Cone pedicles form spatially segregated functionally opposite connections with bipolar cells. Here, we discover that mammalian cones express LRFN2, a cell-adhesion molecule, which localizes pedicle base. LRFN2 stabilizes basal contacts between cell dendrites guide pathway-specific partner choices, encompassing multiple types. In addition, trans-synaptically organizes glutamate receptor clusters, determining contrast preferences pathway. converge in inner retina regulate outputs. We analyze LRFN2’s contributions ON-OFF interactions, pathway asymmetries, neural behavioral responses approaching predators. Our results reveal controls formation vision, supports parallel processing single synapse, shapes coding detection visual threats.

Degeneration of the retina is intrinsically associated with pathogenesis and progression neurodegenerative diseases. However, cellular molecular mechanisms underlying association between neurodegeneration retinal degeneration are still under exploration due to complexity connectivity network nervous system. In this study, RNA-seq data from brains model retinitis pigmentosa (RP) mice previously studied Parkinson's disease (PD) were analyzed explore commonalities degenerative Differentially expressed genes in RP compared disease-related intersecting identified, including Cnr1 Septin14. These verified by quantitative real-time reverse transcription PCR Western blotting experiments. The key proteins CNR1 SEPTIN14 found be potential cotherapeutic targets for disease. conclusion, understanding diseases processes brain will not only facilitate interpretation pathomechanisms but also contribute early diagnosis development new therapeutic strategies.

. Vision restoration approaches, such as prosthetics and optogenetics, provide visual perception to blind individuals in clinical settings. Yet their effectiveness daily life remains a challenge. Stereotyped quantitative tests used trials often fail translate into practical, everyday applications. On the one hand, assessing real-life benefits during is complicated by environmental complexity, reproducibility issues, safety concerns. other predicting behavioral of restorative therapies naturalistic environments may be crucial step before starting minimize patient discomfort unmet expectations.
