Abstract Metastasis is a pivotal event that accelerates the prognosis of cancer patients towards mortality. Therapies aim to induce cell death in metastatic cells require more detailed understanding metastasis for better mitigation. Towards this goal, we discuss details two distinct but overlapping pathways metastasis: classical reversible epithelial-to-mesenchymal transition (hybrid-EMT)-driven transport pathway and an alternative process-driven blebbishield metastatic-witch (BMW) involving process. The knowledge about EMT BMW important therapy cancers as these confer drug resistance coupled immune evasion/suppression. We initially compare it with contexts coordinated oncogenic, metabolic, immunologic, biological events drive metastasis. In particular, how environment apoptosis, ferroptosis, necroptosis, NETosis or recruits cells, fuses it, migrates, permeabilizes vasculature, settles at distant sites establish Finally, therapeutic targets are common both pathways.

Tight junctions (TJ) are cell–cell adhesive structures that define the permeability of barrier-forming epithelia and endothelia. In contrast to this seemingly static function, TJs display a surprisingly high molecular complexity unexpected dynamic regulation, which allows maintain barrier in presence physiological forces response perturbations. Cell–cell adhesion receptors play key roles during regulation TJs. They connect individual cells within cellular sheets link sites contacts underlying actin cytoskeleton. Recent findings support transmitting mechanical promoting phase separation. review, we discuss newly discovered functions cell localized at their role function.

The intestine contains the largest microbial community in human body, gut microbiome. Increasing evidence suggests that it plays a crucial role maintaining overall health. However, while many studies have found correlation between certain diseases and changes microbiome, impact of different compositions on mechanisms by which they contribute to disease are not well understood. Traditional pre-clinical models, such as cell culture or animal limited their ability mimic complexity physiology. New mechanistic organ-on-a-chip, being developed address this issue. These models provide more accurate representation physiology could help bridge gap clinical studies. Gut-on-chip allow researchers better understand underlying effect gut. ABSTRACT Epithelial cells form tissues that generate biological barriers in the body. Tight junctions (TJs) are responsible for maintaining a selectively permeable seal between epithelial cells, but little is known about how TJs dynamically remodel response to physiological forces challenge barrier function, such as cell shape changes (e.g. during division) or tissue stretching developmental morphogenesis). In this Review, we first introduce framework think TJ remodeling across multiple scales: from molecular dynamics, strand cell- and tissue-scale dynamics. We then relate knowledge gained global perturbations of emerging information local events, where transient localized Rho activation actomyosin-mediated contraction promote repair leaks function. conclude by identifying areas field propose ideas future studies address unanswered questions mechanisms drive remodeling.
