For cells to polarize collectively along a tissue plane, asymmetrically localized planar cell polarity (PCP) complexes must form intercellular contacts between neighboring cells. Yet, it is unknown whether asymmetric segregation of PCP requires cell-cell contact, or if autonomous, antagonistic interactions are sufficient for polarization. To test this, we generated mouse chimeras consisting dual PCP-reporter mixed with unlabeled that cannot bridges. In the absence interactions, proteins failed autonomously. Rather, PCP-mediated single interfaces were sort components opposite sides junction, independent junction orientation. Thus, binding critical step initiates sorting opposing generate asymmetry.

Adult mammalian lungs exhibit a fractal pattern, as each successive generation of airways is fraction the size parental branch. Achieving this structure likely requires precise control airway length and diameter, embryonic initially lack scaling observed in adult. In monolayers tubes, directional growth can be regulated by planar cell polarity (PCP) complex. Here, we characterized roles PCP complex components initiation, elongation widening during branching morphogenesis lung. Using tissue-specific knockout mice, surprisingly found that proceeds independently function lung epithelium. Our data thus reveal an explicit independent core complex, suggesting functional diversification vertebrate development. These also essential mesenchyme generating mature

Disruptions in foregut morphogenesis can result life-threatening conditions where the trachea and esophagus fail to separate, such as esophageal atresia (EA) tracheoesophageal fistulas (TEFs). The developmental basis of these congenital anomalies is poorly understood, but recent genome sequencing reveals that de novo variants intracellular trafficking genes are enriched EA/TEF patients. Here, we confirm mutation orthologous Xenopus disrupts trachea-esophageal separation similar Rab11a recycling endosome pathway required localize Vangl-Celsr polarity complexes at luminal cell surface opposite sides tube fuse. Partial loss endosomal or epithelial division orientation. Mutant cells accumulate fusion point, relocalize cadherin, do not separate into distinct esophagus. These data provide insights mechanisms general paradigms tissue during organogenesis.
