Background: Major depressive disorder is a common mental health issue characterized by persistently low mood and high morbidity mortality. The major pathophysiology neuroinflammation, as evidenced elevated cytokine levels. Patients often fail to achieve full remission with the use of currently available antidepressants, prompting search for new treatment options. Schaftoside (SS), flavonoid found in traditional Chinese herbs, has both antioxidant anti-inflammatory properties. However, its antidepressant effects are poorly understood. Methods: Male C57BL/6 mice underwent chronic unpredictable mild stress (CUMS) lipopolysaccharide (LPS) induce depression- anxiety-like behaviors. SS was administered at 40, 80, 160 mg/kg 28 days. effect on depression-like behaviors assessed using behavioral assays, ELISA used measure pro-inflammatory cytokines serum hippocampus. Results: significantly decreased immobility forced swim tail suspension tests, increased sucrose preference test, reduced feeding latency novelty-suppressed test. These findings indicate improved depression showed that lowered interleukin-1 beta (IL-1β), IL-6, tumor necrosis factor-alpha levels hippocampus CUMS mice. Conclusions: Our study indicates anxiolytic effects, possibly through neuroinflammatory processes, making it promising therapeutic candidate depression, thus deserves further investigation into mechanisms clinical efficacy.

BACKGROUND: This study aimed to observe the effect of esketamine on postoperative pain and inflammatory factors in children undergoing tonsillectomy adenoidectomy.METHODS: Eighty scheduled for adenoidectomy were randomly divided into two groups using a random number table: an group (S-ketamine group, N.=40) control (control N.=40). The primary outcome was score, measured Face, Legs, Activity, Cry, Consolability (FLACC) scale score. Secondary outcomes include emergence agitation score (Pediatric Anesthesia Emergence Delirium [PAED]), operation time, recovery postanesthesia care unit (PACU) stay adverse reactions within 48 h after operation, plasma C-reactive protein (CRP) interleukin-6 (IL-6) concentrations.RESULTS: At T1, T2, T3, T4, T5, FLACC scores S-ketamine significantly lower than those (15 min: 2.4±0.7 vs. 3.4±0.8, P<0.01; 1 h: 1.7±0.6 2.2±0.7, 6 1.5±0.6 2.0±0.5, 12 1.8±0.7, P<0.05; 24 1.4±0.6 1.7±0.6, P<0.05). PAED at T1 T2 4.0±0.9 6.4±1.3, 2.1±0.5, P<0.01). Compared with T0, CRP IL-6 concentrations T3 increased (P<0.01), but increase smaller that (IL-6: 111.8±19.2 145.8±22.5, CRP: 1.2±0.5 1.5±0.5, other similar between groups.CONCLUSIONS: Esketamine used pediatric can effectively reduce FLACC, levels without increasing reactions.

Introduction Postoperative delirium (POD) is a common complication after hip fracture surgery in older patients. Esketamine may be beneficial alleviating the occurrence of POD. Our trial aim to investigate whether intravenous administration esketamine can improve POD patients undergoing for fracture. Methods and analysis This randomised controlled will conducted at Chengdu Integrated TCM & Western Medicine Hospital Chengdu, China. A total 260 scheduled under general anaesthesia randomly allocated either an group (group E) or control C) 1:1 ratio (n=130 each group). After tracheal intubation, E receive continuous infusion rate 0.3 mg/kg/h intraoperatively until beginning skin incision closure. Group C equivalent volumes rates 0.9% normal saline; injection methods are accordance with those E. The primary outcome incidence within 3 days surgery, which evaluated using confusion assessment method two times per day. Secondary outcomes subtypes, duration delirium, length hospital stay, pain severity score 30-day all-cause mortality. Pain scores rest numeric rating scale. Abstract Depression, a prevalent psychiatric disorder, exerts severe and debilitating impacts on an individual's mental physical well-being, it is considered chronic illness. Chronic stress plays important role in the pathophysiology of depression. Lactobacillus plantarum Streptococcus thermophilus are psychobiotic bacteria synthesize some neurotransmitters that play pathogenesis In this study, we aimed to investigate therapeutic effects Bactolac ( NBIMCC 8767 + 8258) stress-induced depression rats. Behavioral tests, including sucrose preference test, elevated plus maze forced swim three-chamber sociability were employed assess depressive anxiety-like behaviors.
