The Nile red is a fluorescent and metachromatic dye, for hydrophobic nonpolar materials such as lipids plastics. However, when the microplastics (MP) are contained in natural matrix composed of roots, plant insect fragments rich substances cuticles, chitin, autofluorescence (cell wall, lignin, polyphenols) false fluorescence could generate. In present study, we explore use Methylene blue (C.I. 52015) combination with red, to stain MP an intact organic complex roots rhizome Cyperus papyrus Pontederia sagittata from Floating Treatment Wetlands (FTW) installed at urban eutrophicated ponds Xalapa Veracruz, México. First, sample was stained 0.5 % v/v 0.5% borax reduce nonspecific autofluorescence. After wash distilled water, (1 µg mL-1) ethanol/water. This double reduces background non-plastic materials, obtaining best contrast under green light (ex. 450-490 nm, em. 515 nm). first report easy, fast, non-destructive staining technique detect conditions, that uses together positive staining.
