Abstract: Alzheimer’s disease (AD) is a complex neurological disorder and multiple pathological factors are believed to be involved in the genesis progression of dis-ease. A number hypothesis including Acetylcholinesterase, Monoamine oxidase, β-Amyloid, Tau protein etc. have been proposed for initiation disease. At present, acetylcholine esterase inhibitors memantine (NMDAR antago-nist) only approved therapy symptomatic management AD. Most these single-target drugs miserably failed treatment or halting Multi-factorial diseases like AD require strategies that involve simultaneous modulation network interacting targets. Since last few years, Multi-Target-Directed Ligands (MTDLs) strategy, can simultaneously hit mul-tiple targets, being explored as an effective therapeutic approach In current review article, authors briefly described various pathogenic pathways associated with Importance their design recently reported articles discussed detail.
