ABSTRACT Acute lung injury (ALI) is associated with high morbidity and mortality rates. However, its clinical treatment limited. Currently, the of diseases by regulating microbiota has become a research hotspot. In this study, we investigated protective effects intranasal administration Clostridium butyricum derived extracellular vesicles (EVs) against lipopolysaccharide (LPS)-induced ALI. The results demonstrated that compared LPS group, pre-treatment group C. EVs reduced expression pro-inflammatory cytokines alleviated symptoms in ALI mice inhibiting TLR4/MyD88 signaling pathway. Moreover, inhibited apoptosis-related proteins increased barrier proteins. Additionally, intervention changed composition pulmonary microbiota. At species level, significantly relative abundance Acinetobacter johnsonii , while reversed effect. conclusion, these data demonstrate can prevent LPS-induced reducing inflammation, apoptosis, improving function. regulated to alleviate IMPORTANCE disorder plays an important role progression acute (ALI). very few studies have been conducted treat modulating diversity were altered mice, but ecological balance was restored . study reported mechanism for These reveal importance disease. It provides new approach new-generation probiotics.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline, memory loss, and functional impairments. Despite extensive research, its pathogenesis remains incompletely understood, effective treatments are limited. This study explored the therapeutic potential of agarwood in AD integrating network pharmacology, protein-protein interaction (PPI) analysis, single-cell expression analysis. The results revealed that compounds may modulate key inflammatory genes such as NFKB1, STAT1, TLR4, alleviating neuroinflammation; enhance HSP90 regulate KDR signaling to improve blood-brain barrier (BBB) integrity; promote activity PTPN11 CXCR4 support oligodendrocyte precursor cell (OPC) repair remyelination. Single-cell analysis highlighted cell-type-specific patterns, particularly OPCs endothelial cells, underscoring their relevance pathology. Agarwood's multi-dimensional positions it promising candidate for development novel treatments.
