Abstract: Ferroptosis, a unique form of programmed cell death, is initiated by an excess iron accumulation and lipid peroxidation-induced damage. There growing body evidence indicating that ferroptosis plays critical role in the advancement tumors. The increased metabolic activity higher levels tumor cells make them particularly vulnerable to ferroptosis. As result, targeted induction becoming increasingly promising approach for cancer treatment. This review offers overview regulatory mechanisms ferroptosis, delves into mechanism action traditional small molecule inducers their effects on various In addition, latest progress inducing using new means such as proteolysis-targeting chimeras (PROTACs), photodynamic therapy (PDT), sonodynamic (SDT) nanomaterials summarized. Finally, this discusses challenges opportunities development ferroptosis-inducing agents, focusing discovering targets, improving selectivity, reducing toxic side effects. Targeted protein degradation (TPD) is an emerging therapeutic paradigm aimed at eliminating the disease-causing with aberrant expression. Herein, we report a new approach to inducing intracellular glutathione peroxidase 4 (GPX4) trigger ferroptosis by bridging target heat shock 90 (HSP90), termed HSP90 interactome-mediated proteolysis targeting chimera (HIM-PROTAC). Different series of HIM-PROTACs were synthesized and evaluated, two them, GDCNF-2/GDCNF-11 potently induced via HSP90-mediated ubiquitin-proteasomal GPX4 in HT-1080 cells DC

Elevated levels of the enzyme GPX4 have been detected in tumor tissues, which may play a role cancer progression. We did meta-analysis eight studies encompassing 1180 individuals to evaluate importance cancer, particularly terms prognosis and clinicopathological characteristics. Research results indicate that higher were linked worse overall survival (OS) (HR = 1.47 [95%CI 1.18-1.76], p < .001). lymph node invasion (OR.69 [95% CI.44-1.10], =.12), metastasis (OR 1.58 CI.97-2.55], =.06, <.0001), advanced clinical stage III-IV (OR.82 CI.70-.96], =.001). A sensitivity study revealed general findings constant across all impact intensity. The this suggest increased are not only correlated with reduced rates for patients tumors but it also offers valuable insights regarding traits malignancy metastasis. Based on these connections, has potential serve as biomarker detection, prognosis, targeted therapy.
