Psoriasis, a chronic immune-mediated inflammatory skin disease, influences approximately 2-3% of the world's population. At present, etiology psoriasis remains unclear and there is still no causal treatment available. Recent studies indicate that oxidative stress (OS) T cells dysregulation may participate in pathogenesis psoriasis, among which M1-dominant macrophage polarization crucial contributor. Macrophages mainly polarize into two different subsets, ie, classically activated (M1) alternatively (M2). M1 tends to exacerbate via producing substantial reactive oxygen species (ROS) mediators, encourage OS invasion dysregulation. Thus, targeting can be possible therapeutic alternative for psoriasis. Loganin, belonging iridoid glycosides, pharmaceutically active ingredient originated from Cornus officinalis, exerting multiple biological activities, eg, immunomodulation, antioxidation, anti-inflammation, etc. More importantly, it could effectively suppress polarization, thereby arresting aggression cells' Numerous have confirmed loganin quite reliable diseases suppressing polarization. Psoriasis is a chronic skin disease caused by the interaction of multiple factors that leads to abnormal growth stratum corneum cells and has been called an immortal cancer. Docetaxel trialed for treatment psoriasis due its superior ability induce apoptosis, but insolubility low bioavailability have hampered development. Here, docetaxel (DTX)-loaded liposomes-in-gel (DTX-LP-G) as transdermal delivery was investigated via modulating IL6-HIF-1α-VEGF axis. The results demonstrated DTX-LP-G cumulatively released much higher amount drug into than from DTX-loaded liposomes (DTX-LPs) gel (DTX-G). also most efficient in scavenging hydrogen peroxide free radicals vitro. In mouse model psoriasis, acted preliminary therapeutic agent terms apparent evaluation, splenomegaly, suppression MDA content tissue, down-regulated expression IL6, HIF-1α, VEGF control proliferation vessels, except less pronounced effect on corneum. addition, enrichment analysis can speculate DTX treated resisting production keratin-forming cells.
