Abstract Aromatic natural products are important for improving human health and quality of life. Large-scale availability these compounds from plants is limited by low yield cumbersome extraction. Building high-performance microbial cell factories to produce aromatic means metabolic engineering synthetic biology a viable option. In the context climate change global resource scarcity, choosing solar-powered carbon-fixing cyanobacteria instead chemical heterotrophic microorganisms might be more progressive better solution. this review, we summarize principal strategies production in engineered cyanobacteria, including regulation metabolism, optimization pathway, culture process development, cooperation, highlighting potential microbe as novel chassis cell.

Anti-Neuroinflammatory Coumarins from Paederia Scandens (Lour.) Merr DOI

The aerial parts of Paederia scandens (Lour.) Merr. were extracted with water and the extract was isolated purified through systematic chromatographic separation. Twenty-one coumarins, including six previously undescribed ones, obtained. Their structures elucidated based on comprehensive analyses NMR, HR-ESI-MS, ECD data. anti-neuroinflammatory activities compounds evaluated using an LPS-induced neuroinflammation model in BV-2 microglia cells. Fraxin methyl ether (2) exhibited notable inhibitory effects, IC50 values 17.07 ± 2.88 μM. Moreover, further mechanistic studies compound 2 revealed that it could block nuclear translocation NF- κB reduce expression iNOS, COX-2, IL-1β IL-6, exerting effects. These findings provide a solid foundation for development utilization P. as natural source potential agents.

Previously we discovered that among 15 DNA-binding plant secondary metabolites (PSMs) possessing anticancer activity, 11 compounds cause depletion of the chromatin-bound linker histones H1.2 and/or H1.4. Chromatin remodeling or multiH1 knocking-down is known to promote upregulation repetitive elements, ultimately triggering an interferon (IFN) response. Herein, using HeLa cells and applying fluorescent reporter assay with flow cytometry, immunofluorescence staining quantitative RT-PCR, studied effects PSMs both evicting from chromatin not influencing their location in nucleus. We found (1) 8 PSMs, histone chromatin, activated significantly type I IFN signaling pathway out these resveratrol, berberine, genistein, delphinidin, naringenin curcumin also caused LINE1 expression. Fisetin quercetin, which induced eviction only signaling, but expression; (2) curcumin, sanguinarine kaempferol, causing significant H1.4 presence activate less intensively without any changes (3) four did eviction, displayed neither activation nor enhancement Thus, have shown for first time destabilization observed by accompanied expression often impacts this activation.
