Obesity is a major global health challenge, significantly elevating the risk of cardiovascular disease and type II diabetes. In this study, we identified that self-assembled stearic-acid-modified chitosan oligosaccharide (COA) nanoparticles can efficiently degrade lipid droplets in adipocytes via autolysosomal pathways. By integrating COA with photothermal therapy (PTT), developed combination delivered through an effervescent microneedle (MN) patch for treating diet-induced obesity mice. The MN demonstrated superior skin penetration due to efficient separation needle tips from backing layer. Over four-week treatment period, nanoparticle indocyanine green coloaded patches dramatically reduced body weight white adipose tissue obese mice without affecting their food intake. Histological analysis further revealed droplet size increased expression UCP-1 within tissues vivo. Additionally, improved glucose metabolism insulin sensitivity These findings suggest patch, when combined localized PTT, effectively combat its associated metabolic complications mouse model. This approach presents promising alternative oral injectable loss medications, offering efficacy fewer side effects.
