The spread of antibiotic resistance has become a major concern for public health. As emerging contaminants, various metallic nanoparticles (NPs) and ionic heavy metals have been ubiquitously detected in environments. Although previous studies indicated NPs could exhibit co-selection effects resistance, little is known about whether how they promote via horizontal gene transfer across bacterial genera. This study, we report both CuO copper ions (Cu2+) stimulate the conjugative multiple-drug genes. When exposing bacteria to or Cu2+ at environmental-relevant sub-inhibitory concentrations (e.g., 1-100 μmol/L), conjugation frequencies plasmid-encoded genes genera (i.e., from Escherichia coli Pseudomonas putida) were significantly enhanced (p < 0.05). over-production reactive oxygen species played crucial role promoting transfer. Abstract Antibiotic resistance is a serious global threat for public health. Considering the high abundance of cell-free DNA encoding antibiotic genes (ARGs) in both clinical and environmental settings, natural transformation an important horizontal gene transfer pathway to transmit resistance. It acknowledged that antibiotics are key drivers disseminating resistance, yet contributions non-antibiotic pharmaceuticals on ARGs overlooked. In this study, we report some commonly consumed pharmaceuticals, at clinically environmentally relevant concentrations, significantly facilitated spread through uptake exogenous ARGs. This included nonsteroidal anti-inflammatories, ibuprofen, naproxen, diclofenac, lipid-lowering drug, gemfibrozil, β-blocker propranolol. Abstract Antimicrobial resistance (AMR) poses a worldwide threat to human health and biosecurity. The spread of antibiotic genes (ARGs) via conjugative plasmid transfer is major contributor the evolution this resistance. Although permitted as safe food additives, compounds such saccharine, sucralose, aspartame, acesulfame potassium that are commonly used nonnutritive sweeteners have recently been associated with shifts in gut microbiota similar those caused by antibiotics. The bacterial infection that involves antimicrobial resistance is a rising global threat to public health. Chlorine-based water disinfection processes can inactivate antibiotic resistant bacteria. However, at the same time, these may cause release of genes into as free DNA, and consequently increase risk disseminate via natural transformation. Presently, little known about contribution residual chlorine affecting transformation extracellular (ARGs). This study investigates whether chloramine promote ARGs how this occur. We reveal both chlorine, practically relevant concentrations, significantly stimulated plasmid-encoded by recipient Acinetobacter baylyi ADP1, up 10-fold increase. underlying mechanisms underpinning increased transformations were revealed. Disinfectant exposure induced series cell responses, including levels reactive oxygen species (ROS), membrane damage, ROS-mediated DNA stress response.
