More than 16,000 chemicals are incorporated into plastics to impart properties such as color, flexibility, and durability. These may leach from plastics, resulting in widespread human exposure during everyday use. Two plastic-associated chemicals—bisphenol A (BPA) di(2-ethylhexyl) phthalate (DEHP)—and a class of chemicals—brominated flame retardants [polybrominated diphenyl ethers (PBDEs)]—are credibly linked adverse health cognitive impacts. BPA exposures associated with ischemic heart disease (IHD) stroke, DEHP increased all-cause mortality among persons 55 64 y old, prenatal PBDE mothers IQ losses their children. Phthalic acid esters (PAEs), commonly used as plasticizers, are pervasive in the environment, leading to widespread human exposure. The association between phthalate exposure and metabolic disorders has been increasingly recognized, yet precise biological mechanisms not well-defined. In this study, we explored effects of monoethylhexyl (MEHP) monocyclohexyl (MCHP) on glucose lipid metabolism hepatocytes adipocytes. hepatocytes, MEHP MCHP were observed enhance uptake accumulation a dose-responsive manner, along with upregulating genes involved biosynthesis. Transcriptomic analysis indicated broader impact hepatic gene expression relative MCHP, but particularly promoted gluconeogenesis key enzymes G6PC FBP1. adipocytes, both increased droplet formation, mimicking Peroxisome proliferator-activated receptor γ (PPARγ) agonist rosiglitazone (Rosi). revealed that predominantly altered fatty pathways mature adipocytes (MA), whereas exhibited less impact. Metabolic perturbations from demonstrate shared activation PPARs pathway cell-type discrepancy might be attributed differential PPARγ. Our results indicate disrupt homeostasis liver adipose through involve PPAR adenosine monophosphate-activated protein kinase (AMPK) signaling pathways, highlighting nuanced cellular responses these environmental contaminants.

Population exposure to plastics is increasing, and plasticizers are frequently detected in humans as important ingredients of plastic products. However, patterns harmful their effects on neurofilament light chain (NFL), a marker active brain pathology, currently inconclusive. Herein, we employed range statistical methods thoroughly investigate the impact 24 hazardous varying NFL concentrations blood general population 533 participants. Generalized linear model revealed positive correlation between Mono-isononyl phthalate Mono (2-Ethyl- 5-Hydroxyhexyl) Phthalate (MEHHP) with NFL. Furthermore, significant dose-response relationship was observed MEHHP NFL, while Butyl paraben (Hydroxy-Isononyl) Ester exhibited distinct "inverted U-shaped" nonlinear pattern Additionally, Weighted Quantile Sum allowed us identify mixed all ingredients, Mono(2-Ethyl-5-Oxohexyl) Phthalate, Mono-isobutyl phthalate, Mon butyl Propyl Triclosan occupying prominent positions. Finally, latent profile analysis categorized exposures into high, medium, low patterns, confirming that higher posed risk factor for elevated levels blood. Exposure significantly increases present contributes early detection intervention reduce incidence neurodevelopmental disorder.

The environmental fragmentation of plastics generates a mixture plastic particles various sizes, which frequently co-occur with other mobile and persistent pollutants. Despite the prevalence such scenarios, interaction between micro- nanoplastics (MNPs) their combined effects pollutants, as highly toxic hexavalent chromium (Cr(VI)), remain almost entirely unexplored in mammalian species. This study demonstrated that nanoplastic microplastic co-aggregate together influence Cr bioaccumulation patterns related physiological alterations rats. Following four-week repeated intragastric exposure Wistar rats to MNPs Cr(VI), either alone or combination, significantly enhanced liver, heart, brain, skin. Under co-exposure conditions, Cr(VI) was primary driver cellular observed blood, including shifts immune cell subpopulations (e.g., neutrophils, lymphocytes) red blood indices, while serum biochemistry reflected limited stress. per se decreased creatine kinase activity increased cholesterol levels. In summary, polystyrene increase distribution bioavailability, but does not uniformly exacerbate toxicity. Instead, may selectively alter responses, emphasizing need for deeper understanding potential health risks.
