Microplastics (MPs) are emerging contaminants characterized by persistence, cross-media transport, and complex pollutant interactions, posing serious ecotoxicological risks to ecosystems human health. Effective MPs management requires multi-faced, long-term, strategies involving targeted sampling, quantitative detection, comprehensive risk assessments, all of which entail significant resource investment. Despite advancements in remediation technologies, a holistic governance framework integrating these innovations remains underdeveloped. Transgenerational nanoplastic toxicity could be detected in Caenorhabditis elegans after exposure at the parental generation (P0-G); however, underlying mechanisms remain largely unclear. We aimed to examine role of germline nuclear hormone receptors (NHRs) controlling transgenerational polystyrene nanoparticles (PS-NPs) based on gene expression screening and functional analysis. Among NHR genes, daf-12, nhr-14, nhr-47 expressions were increased nhr-12 was decreased by PS-NPs (1 10 μg/L). alterations these four genes also induced RNAi caused resistance, whereas conferred susceptibility PS-NP toxicity. After exposure, ins-3, daf-28, ins-39 encoding insulin ligands, efn-3 Ephrin ligand, lin-44 Wnt as well their receptor (daf-2, vab-1, and/or mig-1), dysregulated nhr-47, nhr-12.
