Background:Dendrobium officinale glycolipids (DOG), often left as residues after hot water extraction for polysaccharide production, are discarded. Methods: This study investigates the optimal of DOG using response surface methodology, focusing on liquid–solid ratios, ethanol concentrations, temperatures, and times, while preliminarily analyzing DOG’s structural properties. Additionally, differences in antioxidant effects between ascorbic acid based intestinal flora metabolism were further evaluated. Results: The parameters determined follows: ratio 20 mL/g, concentration 70%, temperature 70 °C, time 2.5 h, yielding 2.64 ± 0.18%. In addition, was identified a diglyceride, mainly composed glucose, mannose, linoleic acid, 9,12,15-octadecatrienoic presented certain direct free radicals scavenging effects. animal experiments, unlike increased Bacteroides acidifaciens abundance mice, up-regulated piceatannol expression, down-regulated 1-naphthol which contributed to by enhancing activities SOD GSH-Px reducing MDA content. Conclusions: diglyceride isolated from D. extraction, strong regulating metabolism. These findings could promote efficient utilization support development functional food applications.
