Abstract Background Monitoring labs are a fundamental link in the food safety chain, and regulatory demands competitive economy call for analytical methods that simpler, faster, more rugged, broader scope. The QuEChERSER mega-method introduced 2021 meets these monitoring needs, which includes high sample throughput, automated cleanup of extracts, fast low-pressure gas chromatography (LPGC). Objective goal this work was to extend method additional matrices analytes using LPGC, including comparison performances two different mass spectrometric (MS) analyzers: triple quadrupole tandem MS/MS orbital ion trap (orbitrap) high-resolution (HR)MS. Methods validated 245 pesticides environmental contaminants barley grains hemp pellets instrument top preparation (ITSP) coupled with LPGC-MS/MS or LPGC-HRMS (orbitrap). Results Targeted detection proved be sensitive than orbitrap full data acquisition, leading lower limits quantification (LOQs) yielding acceptable recoveries (70–120%) repeatabilities (RSDs <20%). In barley, 89% compounds met validation criteria 74% HRMS, were 81% 66%, respectively. Qualitatively, HRMS yielded 1% false positives compared 3–4% MS/MS, but due higher LOQs, rates negatives 14–17% vs. 6–10% matrices. Conclusion ITSP+LPGC analysis is robust approach multiple applications. comparison, outperformed terms sensitivity, advantages easier development, greater selectivity, possibility nontargeted/retrospective permit even expansion scope future. Highlights ITSP+LPGC- as part useful efficient way monitor foods.
