Exposure to persistent organic pollutants (POPs), such as dichlorodiphenyltrichloroethane (DDT) and polychlorinated biphenyls (PCBs), has historically been linked population collapses in wildlife. Despite international regulations, these legacy chemicals are still currently detected women of reproductive age, their levels correlate with reduced ovarian reserve, longer time-to-pregnancy, higher risk infertility. However, the specific modes action underlying associations remain unclear. Here, we examined effects five commonly occurring POPs - hexachlorobenzene (HCB), p,p'-dichlorodiphenyldichloroethylene (DDE), 2,3,3',4,4',5-hexachlorobiphenyl (PCB156), 2,2',3,4,4',5,5'-heptachlorobiphenyl (PCB180), perfluorooctane sulfonate (PFOS) mixture on human ovaries vitro. We exposed cancer cell lines COV434, KGN, PA1 well primary cells for 24 h, tissue containing unilaminar follicles 6 days. RNA-sequencing samples concentrations covering epidemiologically relevant revealed significant gene expression changes related central energy metabolism cells, indicating glycolysis, oxidative phosphorylation, fatty acid metabolism, reactive oxygen species potential shared targets POP exposures cells. Alpha-enolase (ENO1), lactate dehydrogenase A (LDHA), cytochrome C oxidase subunit 4I1 (COX4I1), ATP synthase F1 alpha (ATP5A), glutathione peroxidase 4 (GPX4) were validated through qPCR additional culture experiments KGN. In cultures, observed exposure follicle growth atresia protein expression. All exposures, except PCB180, decreased proportion increased atresia. Immunostaining confirmed altered LDHA, ATP5A, GPX4 tissues. Moreover, modified production KGN culture. conclusion, our results demonstrate disruption cellular a novel mode POP-mediated interference ovaries.

There has been emerging research linking per- and poly-fluoroalkyl substances (PFAS) to gamete viability fertility. PFAS, prevalent in the environment water supplies, undergo slow degradation due their C-F bond a long half-life (2.3–8.5 years). In females, PFAS inhibit hypothalamic–pituitary–gonadal (HPG) axis, reducing follicle-stimulating hormone (FSH) luteinizing (LH) levels, leading inhibition of androgen estradiol production. have found cause detrimental effects on egg quality through impairing folliculogenesis. males, can impair sperm motility morphology: two fundamental qualities successful fertilization. exposure proven testosterone production, capacitation, acrosomal reaction. After fertilization, results embryos also investigated, showing reduced development blastocyst stage. The aim this review is report main findings literature impact competency fertilization capability by highlighting key studies both male female We that there significant evidence demonstrating negative impacts fertility after exposure. At high doses, these environmentally abundant widespread compounds significantly affect human

Sperm capacitation is a prerequisite for natural or in vitro fertilization. After capacitation, sperm become hyperactivated and undergo an acrosome reaction, which helps them penetrate the oocyte. Cordycepin, bioactive compound first isolated from Cordyceps militaris, adenosine analog with numerous physiological activities. However, its effects on remain unclear. This study aims to elucidate mechanisms of cordycepin human capacitation.
