Background and Objectives: Age-related macular degeneration (AMD) is the leading cause of blindness, affecting millions worldwide. Its pathogenesis involves death retinal pigment epithelium (RPE), followed by photoreceptor degeneration. Although AMD multifactorial, various genetic markers are strongly associated with disease may serve as biomarkers for evaluating treatment efficacy. This study investigates TAB2 rs237025, IKBKB rs13278372, IKBKG rs2472395 variants their respective serum protein concentrations in relation to occurrence exudative response anti-VEGF treatment. Materials Methods: The case–control involved 961 individuals, they were divided into three groups: control, early AMD, AM patients. Genotyping selected SNPs conducted using a real-time polymerase chain reaction method (RT-PCR). Based on clinical OCT BCVA data, patients categorized one two responders non-responders. data obtained analyzed “IBM SPSS Statistics 29.0” software program. Results: Our revealed that rs237025 allele A was identified risk factor development. same associations remained only females but not males, suggesting gender-specific pathogenetic pathways AMD. Analysis rs13278372 or Lithuanian population no significant associations. On other hand, we found each worse (OR = 0.347; 95% CI: 0.145–0.961; p 0.041). These results suggest potential marker future studies protective variant males only. Also, concentration lower relative control group (median (IQR): 0.442 (0.152) vs. 0.538 (0.337), 0.015). Moreover, who carry GG genotype rs2472394 exhibited significantly reduced compared controls 0.434 (0.199) 0.603 (0.335), 0.012), hypothesis might play role differences Conclusions: both observed distinct pathways. While levels showed association development, treatment, indicating its outcomes. Additionally, be females, particularly rs2472394, progression. findings highlight contribute response.

Ever since the US Food and Drug Administration (FDA) approved first vascular endothelial growth factor (VEGF) antagonist 2 decades ago, inhibitors of VEGF have revolutionized treatment a variety ocular disorders involving pathologic neovascularization retinal exudation. In this perspective, we evaluate current status anti-VEGF therapies real-world challenges encountered with maintaining therapeutic outcomes. Finally, describe novel VEGF-based combinatorial approaches that are in clinical development.

Abstract Uveitis involves a complex interplay of immune cell infiltration and cytokine imbalances, with Th17 cells playing central role in this process. contribute to disease pathogenesis by promoting inflammation, recruiting additional cells, directly damaging retinal tissues. This review discusses the current knowledge on therapeutic strategies targeting Th17-related cytokines, including blockade, small molecule inhibitors, immunomodulatory approaches. Traditionally, cytokines have been viewed as pro-inflammatory agents uveitis. However, emerging research has highlighted capacity response express immunoregulatory notably IL-10, IL-24, TGF-β. suggest that may dualistic includes suppression. In review, we will discuss paradoxical nature regulation inflammation they can both promote mitigate We expected deeper understanding these mechanisms is imperative for innovation novel therapeutics could consider dual By finely tuning preserve integrity function, new treatments bring significant benefits patients aims shed light complexities uveitis its implications future strategies.
