Glial cells play a critical role in primary visual cortex (V1 region) damage caused by optic nerve injury, but the mechanisms driving progression of V1 region injury and glial cell heterogeneity remain poorly understood. This study aimed to investigate changes mice after crush (ONC) single-cell RNA sequencing (scRNA-seq). Hematoxylin eosin (H&E) immunofluorescence staining were used evaluate retinal thickness, astrocytes, microglia ONC mice. Single suspensions prepared analyzed scRNA-seq with Seurat, cellchat, CytoTRACE R software. The expression PTGDS CRYAB was measured qPCR, Western blot, immunofluorescence. After unilateral ONC, thinning both eyes activation astrocytes contralateral observed. Genes related neuroinflammation apoptosis bilateral upregulated, pathways included MAPK, TNF, signaling pathways. Notably, eye exhibited more pronounced differential gene expression, protein neuroinflammation-related genes Ptgds Cryab increased. We further investigated pseudotime trajectories microglia, demonstrating key branches that dominate neuroinflammation. generates an atlas mouse brain, highlighting suggesting as potential targets reduce
