Moringa oleifera Lam., a perennial species of the Moringaceae family, is esteemed for its multifaceted nutritional, medicinal, and economic properties. M. leaves are abundant in bioactive compounds with potential health benefits, yet existing structural functional studies these bioactives remain insufficiently comprehensive. This study aimed to isolate active compound from investigate mechanism inhibiting lipid absorption as pancreatic lipase inhibitor. In present study, octacosanol (OCT) was isolated purified leaves, demonstrating notable inhibitory activity an IC50 7.87 ± 0.72 μg/mL, effectively suppressing uptake HepG2 cells. Simulated digestion assays indicated that OCT retained 73.5% activity, recorded inhibition rate 63.62%. Molecular docking analyses revealed binds affinity comparable orlistat stronger than 4-nitrophenyl palmitate. The binding key sites (Ser152, His263, Asp176) likely disrupts enzyme's conformation, decreasing substrate affinity. Additionally, significantly attenuated synthesis total cholesterol triglycerides. elucidates lipid-lowering provides theoretical foundation application food production biomedicine.
