Background: Heavy metals can cause serious health problems that affect different organs. Cadmium (Cd) is an environmental contaminant known for its toxicological consequences on Hepatotoxicity a effect of exposure to Cd with oxidative stress (OS) and inflammation playing central role. Diallyl disulfide (DADS), organo-sulfur compound found in garlic, cytoprotective antioxidant effects. In this study, the DADS Cd-induced inflammation, liver injury was investigated. Methods: supplemented 14 days via oral gavage, single intraperitoneal dose (1.2 mg/kg body weight) administered rats day 7. Blood samples were collected at end experiment analyses. Results: administration resulted remarkable hepatic dysfunction, degenerative changes, necrosis, infiltration inflammatory cells, collagen deposition other histopathological alterations. increased malondialdehyde (MDA) nitric oxide (NO) (p < 0.001), upregulated toll-like receptor (TLR)-4, nuclear factor-kappaB (NF-κB), pro-inflammatory mediators, caspase-3 0.001) whereas decreased glutathione (GSH) enzymes 0.001). downregulated peroxisome proliferator activated gamma (PPARγ), transcription factor involved OS suppression ameliorated tissue alterations, attenuated apoptosis, suppressed TLR-4/NF-κB signaling, enhanced antioxidants. addition, PPARγ Cd-administered rats. Conclusions: effective against hepatotoxicity beneficial effects are linked apoptosis upregulation PPARγ. could be valuable protect individuals risk exposure, pending further studies elucidate underlying mechanism(s).
