Abstract Advances in food production systems and customer acceptance have led to the commercial launch of dietary proteins produced via modern biotechnological approaches as alternatives traditional agricultural sources. At same time, a deeper understanding how components interact with gut microbiome has highlighted importance nuances underpinning diet-microbiome interactions. Novel distinct post-translational modifications resulting from their respective not been characterized, nor they may differ traditionally counterparts. We found that despite strong similarities composition, significantly differs between these sources, reflecting biosynthetic machinery systems. Three novel endo-β-N-acetylglucosaminidases (AVUL01, BCAC01, and BFIN01) classified as members of the glucoside hydrolase (GH) family 18 were identified from human fecal samples then cloned characterized for their ability to hydrolyze two distinct classes N-glycans. Endo-β-N-acetylglucosaminidases (ENGases) are known hydrolysis chitin N,N′-diacetylchitobiose core N-linked glycans, depending on glycan architecture. N-glycans have shown bioactivity substrates in gut microbiome microbes that encode ENGases, thus demonstrating ecological relevance gut. However, types N-glycan structures, example, oligomannosidic or complex, been enrich different within Novel advances food technology commercialized animal-derived dietary proteins with instead traditionally complex using precision fermentation. This indicates there is an unmet need identify gut-derived ENGases act upon determine whether these alter ecology. AVUL01, BFIN01 all demonstrated activity exclusively RNase B bovine lactoferrin; however, they failed show α-1,3-core fucosylated high-mannose derived fetuin horseradish peroxidase, respectively. These results suggest α-1,3 fucosylation architecture inhibit BFIN01. Furthermore, performed significantly better than resulting a greater amount N-glycans, suggesting certain may possess enhanced specificity kinetics evolutionary strategy compete resources.

Introduction N- trans -feruloyltyramine (NFT) is a bioactive compound present in many plant sources. The purpose of the studies was to investigate adverse effects, if any, NFT produced through precision fermentation. Methods An vitro Ames test performed with using bacterial strains at concentrations up 1580 µg/plate and without S9. micronucleus assay human peripheral blood cells culture, without, metabolic activation three different doses. In subchronic toxicity study, adult Sprague Dawley rats (10/sex/group) were fed diets prepared target doses 0, 5000, 10,000 or 20,000 ppm for 90 days. Results assay, there no NFT-related concentration dependent increases revertant colony numbers any tester strains. statistically significant increase number binucleated micronuclei compared vehicle control. found be non-genotoxic when evaluated assays. 90-day rodent well tolerated, related findings observed dose levels tested. There histopathological changes high group both sexes. Conclusion No effect level determined as 1474 mg/kg body weight/day males 1958 females based on actual intake tested under experimental conditions employed.

Objectives: This study investigated the effects of 2-week ingestion hemp fiber (high and low doses) versus placebo bars on gut permeability plasma metabolite shifts during recovery from 2.25 h intensive cycling. Hemp hull powder is a rich source two bioactive compounds, N-trans-caffeoyl tyramine (NCT) N-trans-feruloyl (NFT), with potential health benefits. Methods: The participants included 23 male female cyclists. A three-arm randomized, placebo-controlled, double-blind, crossover design was used supplementation periods washout periods. Supplement provided 20, 5, or 0 g/d powder. Participants engaged in an cycling bout at end each three Five blood samples were collected before after (overnight fasted state), h-, 1.5 3 h-post-exercise. Five-hour urine pre-supplementation post-2.25 ingesting sugar solution containing 5 g lactulose, 100 mg 13C mannitol, 1.9 mannitol 450 mL water. An increase post-exercise lactulose/13C ratio (L:13CM) as primary indicator altered permeability. Other outcome measures muscle damage biomarkers (serum creatine kinase, myoglobin), serum cortisol, complete cell counts, metabolites using untargeted metabolomics. Results: No trial differences found for L:13CM, biomarkers. Orthogonal partial least-squares discriminant analysis (OPLSDA) showed distinct when comparing high- low-dose compared to (R2Y = 0.987 0.995, respectively). Variable Importance Projection (VIP) scores identified several relevant metabolites, including 3-hydroxy-4-methoxybenzoic acid (VIP 1.9), serotonin 1.5), 5-hydroxytryptophan 1.4), 4-methoxycinnamic 1.4). Mummichog significant intake multiple metabolic pathways, alpha-linolenic acid, porphyrin, sphingolipid, arginine proline, tryptophan, bile metabolism. Conclusions: period did not have effect cyclists (2.25 bout) data. On contrary, metabolomics that combination consuming nutrient-rich exercising 135 min increased levels beneficial those derived healthy
