Sarcoplasmic calcium-binding protein identified in Crassostrea angulata (Cra a 4), is stable allergen; the methods to reduce its allergenicity are still limited. The present study aimed develop hypoallergenic enzymatic hydrolyzate of Cra 4 (ECra 4) by enzymolysis. After screening proteases bioinformatics and optimization enzymolysis conditions, ECra was obtained with Trypsin. Further, used stimulate splenocytes from 4-sensitized mice, findings that could CD4+ T-cell proliferation, inhibit IL-4 secretion, promote TGF-β secretion. Spectroscopy analysis revealed destroyed structure 4, which affected allergenicity. Peptidomic B-cell epitopes while retaining epitopes. Overall, hypoallergenicity stimulating ability can be as an immune tolerance product for further study.

Shrimp are highly favored by consumers for their delicious taste and rich nutritional value. However, reports of allergic reactions caused shrimp its derivatives have been increasing, significantly impacting consumer health posing a growing global food safety concern. This article introduces the structure biochemical characteristics major allergenic proteins in shrimp, including tropomyosin (TM), arginine kinase, sarcoplasmic calcium-binding protein, myosin light chain, troponin C, hemocyanin. Currently, there is no effective treatment allergies, prevention mainly achieved avoiding consumption. The study allergen sensitization reduction technology great significance to development hypoallergenic or desensitized products. provides detailed overview effects common processing techniques, physical, chemical, biological, combined methods, on allergenicity allergens; instance, binding rate immunoglobulin E (IgE) was reduced 73.59% after treating TM with high pressure (500 MPa) at 55 °C 10 min recognition IgE decreased 89.4% average pepsin (30 μg/mL, pH 2) 2 h. These techniques provide references aquatic products foods.

Conjugation of enzymes to lipid membranes is a key approach reconstitute fascinating features cell organelles and deduce the nature membrane-involved biological events. In this work, papain was covalently conjugated via cross-linker phospholipid vesicles (liposomes) tethered with poly(ethylene glycol) (PEG) at 25 °C pH = 7.0, followed by chromatographic purification. The size PEG moiety type were optimized obtain PEG-tethered liposome-conjugated (liposome-PEG-papain). Slight conformational changes membrane-conjugated in both its secondary tertiary structures revealed using circular dichroism intrinsic fluorescence measurements. Notably, heat treatment liposome-PEG-papain dispersion 77 or 84 caused permeabilization 5(6)-carboxyfluorescein. Furthermore, exhibited digestion activity casein 37 7.6. structural flexibility liposomes as enzyme carriers may provide opportunity functionalize biomacromolecules.
