Triploid rainbow trout is a hypoxia-sensitive fish species, and susceptible to death in summer with high water temperature induced acute hypoxia. The study was conducted evaluate the effect of protein diet carbohydrate on hypoxic tolerance triploid trout. Two isolipidic isoenergetic diets low protein-high level (LPHC) protein-low (HPLC) were specifically formulated. After feeding trial 80 days, two group treated challenge. Plasma biochemical parameters, hepatic anti-stress, antioxidant immune related genes expression, glucose, lipid, amino acid metabolism expression then analyzed. results showed that compared HPLC diet, LPHC could result excessive plasma levels alanine aminotransferase (ALT; 36.3 VS 14.0 U/L), aspartate (AST; 978 366 alkaline phosphatase (ALP; 374 204 creatine phosphokinase (CPK; 7538 4937 U/L) lactate dehydrogenase (LDH; 940 451 U/L). In addition, remodel lipid based mRNA regulating glycometabolism, lipometabolism, acids metabolism. also enhanced anti-stress (HSP70, HIF-1a HIF-2a), (Nrf2, SOD, CAT, GPx HO-1) (IFN-γ, IL-1β, TNF-α IL-8). total, aggravate stress trout, while improve tolerance.

Introduction Parasite-mediated selection is considered one of the potential mechanisms contributing to coexistence asexual-sexual complexes. Gibel carp ( Carassius gibelio ), an invasive fish species in Europe, often forms populations composed gynogenetic and sexual specimens. Methods The experimental infection was induced gibel using eye-fluke Diplostomum pseudospathaceum (Trematoda), transcriptome profile spleen as a major immune organ analyzed reveal differentially expressed immunity-associated genes related D. differing between carp. Results High parasite found when compared genetically diverse sexuals. Although metacercariae are situated immune-privileged organ, our results show that eye trematodes may induce host response. We differential gene expression by infection, with various impacts on hosts, documenting for majority DEGs upregulation sexuals, downregulation asexuals. Differences regulation were evidenced many genes. GO analyses revealed importance assigned terms: function, Notch signaling pathway, MAP kinase tyrosine/threonine/phosphatase activity, chemokine receptor activity. KEGG involved 12 pathways – specifically, FoxO signaling, adipocytokine TGF-beta apoptosis, C-type lectin efferocytosis, intestinal network IgA production, insulin virion - human immunodeficiency virus, Toll-like phosphatidylinositol system. Discussion Our study indicates limited asexual cope higher (likely loss capacity effective response) highlights important role molecular associated immunity carp, potentially its invasiveness.

Phosphatidylethanolamine (PE) is a ubiquitous bioactive lipid in cells, which participates regulating many metabolic processes. Exogenous PE has been reported to play positive regulatory role macrophage inflammatory responses. However, the molecular mechanisms of inflammation are not completely understood. In present study, transcriptomic analysis PE-stimulated macrophages large yellow croaker revealed that differentially expressed genes were mainly active cellular components mitochondrial respiratory chain, corresponded significant enrichment oxidative phosphorylation pathway. Consistent with this result, significantly increased ATP content and protein expression NDUFB3 (mitochondrial chain complex I subunit) macrophages. Meanwhile, data showed treatment downregulated transcript levels nlrp3 upregulated suppressor cytokine signaling 3 (socs3), suggesting may alleviate by interfering activation NLRP3 inflammasome. Further attenuated dietary PA-mediated via NLRP3-Caspase-1 vitro vivo. Given abundance strongly correlated function, study hypothesized PE-mediated modulation be attributed effects on function. As expected, ameliorated PA-induced dysfunction reduced intracellular reactive oxygen species production malondialdehyde macrophages, indicating improvement function an important mechanism involved effect inflammation. conclusion, elucidates critical regulation expands understanding phospholipid metabolism fatty acid-induced This provide new intervention targets nutritional strategies for improving chronic diseases.

Hypoxia triggers stress, leading to significant alterations in gene expression patterns, which turn affect fish's growth and development. Real-time quantitative PCR (RT-qPCR) is a pivotal technique for assessing changes expression. However, its accuracy highly contingent upon the stable of reference genes. Ribosomal RNA (18s), β-actin (actb), elongation factor 1-α (ef1a), α tubulin (tuba), ribosomal protein L17 (rpl17) are widely used genes, but their stability tissues black rockfish under hypoxic conditions remains unclear. The genes was detected by RT-qPCR assessed Delta Ct, geNorm, NormFinder, BestKeeper algorithms. Results showed that tuba exhibited liver, heart, gill normoxic conditions, liver head kidney conditions. Ef1a identified as most stably expressed tissue hypoxia. For heart studies, rpl17 were recommended 18s high spleen Actb tissue-specific stability, it necessary select appropriate based on specific type studies These findings help enhancing analysis mechanism hypoxia rockfish.

Erythropoiesis is the process formation new erythrocytes, from hemopoietic stem cells, through differentiation erythroblasts, maturation cells. This step‐wise governed by complex transcriptional, post‐translational, epigenetic programs response extracellular signals. review summarizes developing knowledge regarding teleosts, including wave site erythropoiesis, specification erythroid lineage, molecular regulation mechanisms.
