Hydroxychloroquine (HCQ) is an important public health therapeutic agent widely used in the prevention and treatment of malaria autoimmune diseases, with some antiviral effects, as well a common autophagy inhibitor. Its autophagy-inhibiting effect attracts great research interest mammals but still little studied fish. We even have no idea about effects HCQ on different tissues fish what concentrations should be for safety studies. This study investigated concentration treatments survival, tissue structure, expressions genes related to autophagy, apoptosis, inflammation gill, spleen, testis, ovary Sebastiscus marmoratus . The results showed that higher (77.40 154.80 mg/mL) led mass mortality within hour. half-lethal ( LC 50 , 24 h) S. was approximately 48.95 mg/mL. When exposed 30.96 mg/mL hours, blocked revealed by electron microscopy from spleen testis autophagy-related LC3 / Cx43 ), apoptosis-related Cas3 p53 inflammation-related TNF-α IL8 ) exhibited tissue-specific dose-dependent responses. gonads preferential expression all these were found sensitive regular after treatment. For example, at mg/mL, demonstrated regularity suggests it ideal candidate studying role or autophagy. systematically response provided optional assay key tissues, serving reference future studies Furthermore, potential crosstalk between inflammatory pathways initially identified this could helpful regulation marine

Abstract An in vivo mouse model of Clonorchis sinensis (C. sinensis) infection with or without the administration autophagy inhibitor chloroquine (CQ) stimulation was established to assess possible involvement autophagic response during C. infection. Abnormal liver function observed at four, six, eight weeks post-infection, as indicated by elevated levels ALT/GPT, AST/GOT, TBIL, α-SMA infected group. Our findings activated autophagy, shown a decreased LC3II/I ratio and accumulated P62 expression mice. Interestingly, CQ exhibited dual opposing effects In early stage infection, engagement appeared mitigates symptoms reducing inflammation fibrotic responses. However, later might contribute parasite survival evading targeting, thereby exacerbating hepatic impairment worsening fibrosis. Autophagy suppressed throughout These observations attested that triggered highlighted complex role for CQ, both protective detrimental effects, progression
