The roles of (p)ppGpp in regulating cytosolic proteins are well established; however, their effects on membrane remodeling remain elusive. translocation signal recognition particle (SRP)-dependent can be modulated through binding to two key GTPase components: FtsY, which interacts with SecYEG, and Ffh, a homolog SRP54. A (p)ppGpp-specific Broccoli RNA aptamer the chemometer PyDPA were used quantify levels ΔrelA ΔrelA/ΔspoT strains Acinetobacter baumannii, confirming stepwise reduction following order: wild-type > ΔrelA/ΔspoT. strain, intermediate levels, exhibited increased outer vesicle (OMV) production, whereas strain reduced highlighting non-linear relationship between OMV production levels. Scanning transmission electron microscopy revealed (p)ppGpp-dependent changes cell envelope integrity: relA mutant disruption leading formation, relA/spoT maintained an intact membrane, suggesting that stability is by fine-tuned Western blotting proteomic analyses identified significant OmpA accumulation inner SRP-dependent proteins, including NuoB, NuoL, TolA, strain. These findings indicate crucial for protein incorporation A. baumannii. Regulation using CRISPRi system formation peaked at concentrations, importance precise adjustment bacterial phenotypes.
