Abstract The lipid mediator platelet-activating factor (PAF) and its receptor (PAFR) signaling play critical roles in a wide range of physiological pathophysiological conditions, including cancer growth metastasis. ability PAFR to interact with other oncogenic cascades makes it promising target for treatment. Moreover, numerous natural synthetic compounds, characterized by diverse pharmacological activities such as anti-inflammatory anti-tumor effects, have been explored their potential PAF antagonists. In this review, we provide comprehensive evidence regarding the PAF/PAFR pathway, highlighting effectiveness various classes inhibitors antagonists across multiple models. Notably, synergistic effects enhancing efficacy chemotherapy radiation therapy several experimental models are also discussed. Overall, synthesis literature review indicates that targeting axis represents approach treatment exerts synergy therapy.
